Crenolanib population pharmacokinetics and covariate influence were characterized in this patient population.
Methods
Patients enrolled on this phase I study (NCT01393912) received oral crenolanib once daily. Serial single-dose and steady-state serum pharmacokinetic samples were collected and analyzed using a validated LC-ESI-MS/MS method. Population modeling and covariate analysis evaluating demographics, laboratory values, and comedications were performed. The impact of significant covariates on crenolanib exposure was further explored using model simulations.
Results
Crenolanib serum concentrations were analyzed for 5</pubmed_abstract><journal>Cancer chemotherapy and pharmacology</journal><pubmed_title>Population pharmacokinetics of crenolanib in children and young adults with brain tumors.</pubmed_title><pmcid>PMC8957602</pmcid><funding_grant_id>P30 CA021765</funding_grant_id><funding_grant_id>R25CA23944</funding_grant_id><funding_grant_id>R25 CA023944</funding_grant_id><funding_grant_id>R01CA154619</funding_grant_id><funding_grant_id>CA21765</funding_grant_id><pubmed_authors>Bisbee C</pubmed_authors><pubmed_authors>Campagne O</pubmed_authors><pubmed_authors>Gajjar A</pubmed_authors><pubmed_authors>Tinkle CL</pubmed_authors><pubmed_authors>Stewart CF</pubmed_authors></additional><is_claimable>false</is_claimable>

Purpose
Crenolanib, an oral inhibitor of platelet-derived growth factor receptor, was evaluated to treat children and young adults with brain tumors.
