Immunotherapy strategies are attractive given the relatively intact immune system of patients with LGG and the slow tumor growth rate. However, accumulation of the oncometabolite D-2-hydroxyglutarate (D-2HG) in IDH-mutated gliomas leads to suppression of inflammatory pathways in the tumor microenvironment, thereby contributing to the 'cold' tumor phenotype. Inhibiting D-2HG production presents an opportunity to generate a robust antitumor response following tumor antigen vaccination and immune checkpoint blockade.
Methods
An IDH1R132H glioma model was created in sy</pubmed_abstract><journal>Journal for immunotherapy of cancer</journal><pubmed_title>Inhibition of D-2HG leads to upregulation of a proinflammatory gene signature in a novel HLA-A2/HLA-DR1 transgenic mouse model of IDH1R132H-expressing glioma.</pubmed_title><pmcid>PMC9174833</pmcid><funding_grant_id>R01 CA222965</funding_grant_id><funding_grant_id>R35 NS105068</funding_grant_id><funding_grant_id>T32 CA151022</funding_grant_id><pubmed_authors>Mende AL</pubmed_authors><pubmed_authors>Okada H</pubmed_authors><pubmed_authors>Hudson C</pubmed_authors><pubmed_authors>Tron AE</pubmed_authors><pubmed_authors>Narayanaswamy R</pubmed_authors><pubmed_authors>Nejo T</pubmed_authors><pubmed_authors>Yamamichi A</pubmed_authors><pubmed_authors>Diebold D</pubmed_authors><pubmed_authors>Montoya M</pubmed_authors><pubmed_authors>Lu M</pubmed_authors><pubmed_authors>Ronseaux S</pubmed_authors><pubmed_authors>Chuntova P</pubmed_authors><pubmed_authors>Nicolay B</pubmed_authors><pubmed_authors>Downey KM</pubmed_authors><pubmed_authors>Chen T</pubmed_authors><pubmed_authors>Hyer ML</pubmed_authors></additional><is_claimable>false</is_claimable>

Background
Long-term prognosis of WHO grade II, isocitrate dehydrogenase (IDH)-mutated low-grade glioma (LGG) is poor due to high risks of recurrence and malignant transformation into high-grade glioma.
