Here we describe a detailed protocol of pressure cycling technology (PCT)-assisted sample preparation for proteomic analysis of biopsy tissues. A piece of fresh frozen or formalin-fixed paraffin-embedded tissue weighing ~0.1-2 mg is placed in a 150 μL pressure-resistant tube called a PCT-MicroTube with proper lysis buffer. After closing with a PCT-MicroPestle, a batch of 16 PCT-MicroTubes are placed in a Barocycler, which imposes oscillating pressure to the samples from one atmosphere to up to ~3,000 times atmospheric pressure. The pressure cycling schemes are optimized for tissue lysis and protein digestion, and can be programmed in the Barocycler to allow reprod</pubmed_abstract><journal>Nature protocols</journal><pagination>2307-2325</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC9362583</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>High-throughput proteomic sample preparation using pressure cycling technology.</pubmed_title><pmcid>PMC9362583</pmcid><pubmed_authors>Xu L</pubmed_authors><pubmed_authors>Yi X</pubmed_authors><pubmed_authors>Zhu Y</pubmed_authors><pubmed_authors>Wu C</pubmed_authors><pubmed_authors>Yue L</pubmed_authors><pubmed_authors>Guo T</pubmed_authors><pubmed_authors>Gao H</pubmed_authors><pubmed_authors>Xue Z</pubmed_authors><pubmed_authors>Chen C</pubmed_authors><pubmed_authors>Yu J</pubmed_authors><pubmed_authors>Liu W</pubmed_authors><pubmed_authors>Ge W</pubmed_authors><pubmed_authors>Sun R</pubmed_authors><pubmed_authors>Qian L</pubmed_authors><pubmed_authors>Cai X</pubmed_authors></additional><is_claimable>false</is_claimable><name>High-throughput proteomic sample preparation using pressure cycling technology.</name><description>High-throughput lysis and proteolytic digestion of biopsy-level tissue specimens is a major bottleneck for clinical proteomics.
