Herein, we conducted a systematic comparative analysis of gene expression/progression-free survival relationship from two phase 3 trials (PALOMA-2 [first-line] and PALOMA-3 [≥second-line]). In the ET-only arm, there was no inter-therapy line correlation. However, adding palbociclib resulted in concordant biomarkers independent of initial ET responsiveness, with shared sensitivity genes enriched in estrogen response and resistance genes over-represented</pubmed_abstract><journal>NPJ precision oncology</journal><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC9381541</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Comparative biomarker analysis of PALOMA-2/3 trials for palbociclib</pubmed_title><pmcid>PMC9381541</pmcid><pubmed_authors>Gelmon K</pubmed_authors><pubmed_authors>Harbeck N</pubmed_authors><pubmed_authors>Zhu Z</pubmed_authors><pubmed_authors>Cristofanilli M</pubmed_authors><pubmed_authors>Cao J</pubmed_authors><pubmed_authors>Rejto P</pubmed_authors><pubmed_authors>Lu D</pubmed_authors><pubmed_authors>VanArsdale T</pubmed_authors><pubmed_authors>Andre F</pubmed_authors><pubmed_authors>Dieras V</pubmed_authors><pubmed_authors>Liu Y</pubmed_authors><pubmed_authors>Wei P</pubmed_authors><pubmed_authors>Yan Z</pubmed_authors><pubmed_authors>Huang X</pubmed_authors><pubmed_authors>Turner N</pubmed_authors><pubmed_authors>Loibl S</pubmed_authors><pubmed_authors>Loi S</pubmed_authors><pubmed_authors>Martin M</pubmed_authors><pubmed_authors>Finn R</pubmed_authors><pubmed_authors>Zhang C</pubmed_authors><pubmed_authors>Rugo H</pubmed_authors></additional><is_claimable>false</is_claimable><name>Comparative biomarker analysis of PALOMA-2/3 trials for palbociclib</name><description>While cyclin-dependent kinase 4/6 (CDK4/6) inhibitors, including palbociclib, combined with endocrine therapy (ET), are becoming the standard-of-care for hormone receptor–positive/human epidermal growth factor receptor 2‒negative metastatic breast cancer, further mechanistic insights are needed to maximize benefit from the treatment regimen.
