Seven distinct LASV lineages circulate through western Africa, among which lineage I (LI), the first to be identified, is particularly resistant to antibody neutralization. Lineage I LASV evades neutralization by half of known antibodies in the GPC-A antibody competition group and all but one of the antibodies in the GPC-B competition group. Here, we solve two cryo-electron microscopy (cryo-EM) structures of LI GP in complex with a GPC-A and a GPC-B antibody. We used complementary structural and biochemical techniques to identify single-amino-acid substitutions in LI that are responsible for immune evasion by each antibody group. Further, we show that LI infection is more dependent on the endosomal receptor lysosome-</pubmed_abstract><journal>mBio</journal><pubmed_title>Neutralizing Antibodies against Lassa Virus Lineage I.</pubmed_title><pmcid>PMC9426445</pmcid><funding_grant_id>R21 AI137809</funding_grant_id><funding_grant_id>U19 AI142790</funding_grant_id><funding_grant_id>R01 AI141251</funding_grant_id><funding_grant_id>T32 AI125179</funding_grant_id><funding_grant_id>R01AI141251</funding_grant_id><pubmed_authors>Li H</pubmed_authors><pubmed_authors>Harkins SS</pubmed_authors><pubmed_authors>Zandonatti MA</pubmed_authors><pubmed_authors>Moon-Walker A</pubmed_authors><pubmed_authors>Robinson JE</pubmed_authors><pubmed_authors>Garry RF</pubmed_authors><pubmed_authors>Buck TK</pubmed_authors><pubmed_authors>Branco LM</pubmed_authors><pubmed_authors>Hastie KM</pubmed_authors><pubmed_authors>Schendel SL</pubmed_authors><pubmed_authors>Enriquez AS</pubmed_authors><pubmed_authors>Saphire EO</pubmed_authors></additional><is_claimable>false</is_claimable><name>Neutralizing Antibodies against Lassa Virus Lineage I.</name><description>Lassa virus (LASV) is the causative agent of the deadly Lassa fever (LF).
