Previous studies have suggested an association between CD151 and distinct consequences in osteosarcoma tumorigenicity. However, the potential of CD151 as a therapeutic target has not yet been sufficiently explored. Here, we performed integrated transcriptomic and metabolomic analyses of osteosarcoma and identified sphingolipid metabolism as the top CD151-regulated pathway. CD151 regulates sphingolipid metabolism primarily through SPTCL1, the first rate-limiting enzyme in sphingolipid biosynthesis. Mechanistically, depletion of CD151 enhanced c-myc polyubiquitination and subsequent degradation. c-myc is vital for the transcriptional activation of SPTLC</pubmed_abstract><journal>Cell &amp; bioscience</journal><pubmed_title>Inhibition of sphingolipid metabolism in osteosarcoma protects against CD151-mediated tumorigenicity.</pubmed_title><pmcid>PMC9548188</pmcid><funding_grant_id>81902734</funding_grant_id><funding_grant_id>82072969</funding_grant_id><funding_grant_id>81772859</funding_grant_id><pubmed_authors>Hua Y</pubmed_authors><pubmed_authors>Jin X</pubmed_authors><pubmed_authors>Shen J</pubmed_authors><pubmed_authors>Wang C</pubmed_authors><pubmed_authors>Ma X</pubmed_authors><pubmed_authors>Wang H</pubmed_authors><pubmed_authors>Sun W</pubmed_authors><pubmed_authors>Xu J</pubmed_authors><pubmed_authors>Sun M</pubmed_authors><pubmed_authors>Zhang T</pubmed_authors><pubmed_authors>Zhang Y</pubmed_authors><pubmed_authors>Zan P</pubmed_authors><pubmed_authors>Wang Z</pubmed_authors></additional><is_claimable>false</is_claimable><name>Inhibition of sphingolipid metabolism in osteosarcoma protects against CD151-mediated tumorigenicity.</name><description>Osteosarcoma is the most common primary bone tumor, with a poor prognosis owing to the lack of efficient molecular-based targeted therapies.
