As a member of the basic helix-loop-helix PER-ARNT-SIM (bHLH-PAS) transcription factor family, NPAS4 is distinguished by an ability to form functional heterodimers with aryl hydrocarbon receptor nuclear translocator (ARNT) and ARNT2, both of which are also bHLH-PAS family members. Here, we describe the quaternary architectures of NPAS4-ARNT and NPAS4-ARNT2 heterodimers in complexes involving DNA response elements. Our crystallographic studies reveal a uniquely interconnected domain conformation for the NPAS4 protein itself, as well as its differentially configured heterodimeric arrangements with both ARNT a</pubmed_abstract><journal>Proceedings of the National Academy of Sciences of the United States of America</journal><pubmed_title>Structures of NPAS4-ARNT and NPAS4-ARNT2 heterodimers reveal new dimerization modalities in the bHLH-PAS transcription factor family.</pubmed_title><pmcid>PMC9674253</pmcid><funding_grant_id>tsqn201909004</funding_grant_id><funding_grant_id>ZR2021JQ30</funding_grant_id><funding_grant_id>2020QNQT009</funding_grant_id><funding_grant_id>210664/Z/18/Z</funding_grant_id><funding_grant_id>BK20170399</funding_grant_id><funding_grant_id>22177063</funding_grant_id><pubmed_authors>Jing L</pubmed_authors><pubmed_authors>Diao X</pubmed_authors><pubmed_authors>Li F</pubmed_authors><pubmed_authors>Rastinejad F</pubmed_authors><pubmed_authors>Zhang M</pubmed_authors><pubmed_authors>Wu D</pubmed_authors><pubmed_authors>Sun X</pubmed_authors><pubmed_authors>Zhuang J</pubmed_authors></additional><is_claimable>false</is_claimable><name>Structures of NPAS4-ARNT and NPAS4-ARNT2 heterodimers reveal new dimerization modalities in the bHLH-PAS transcription factor family.</name><description>Neuronal PER-ARNT-SIM (PAS) domain protein 4 (NPAS4) is a protective transcriptional regulator whose dysfunction has been linked to a variety of neuropsychiatric and metabolic diseases.
