Here, we describe our hit-to-lead efforts in search of a selective inhibitor of human Polθ (encoded by POLQ). A high-throughput screening campaign of 350,000 compounds identified an 11 micromolar hit, giving rise to the N2-substituted fused pyrazolo series, which was validated by biophysical methods. Structure-based drug design efforts along with optimization of cellular potency and ADME ultimately led to the identification of <b>RP-6685</b>: a potent, selective, and orally bioavailable Polθ inhibitor that showed in vivo efficacy in an HCT116 BRCA2-/- mouse tumor xenograft model.</pubmed_abstract><journal>Journal of medicinal chemistry</journal><pubmed_title>Identification of <b>RP-6685</b>, an Orally Bioavailable Compound that Inhibits the DNA Polymerase Activity of Polθ.</pubmed_title><pmcid>PMC9942948</pmcid><funding_grant_id>PJT-178026</funding_grant_id><funding_grant_id>P30 CA008748</funding_grant_id><funding_grant_id>P30 GM124165</funding_grant_id><funding_grant_id>FDN-143277</funding_grant_id><funding_grant_id>S10 OD021527</funding_grant_id><funding_grant_id>PJT-180338</funding_grant_id><pubmed_authors>Martino G</pubmed_authors><pubmed_authors>Casas-Selves M</pubmed_authors><pubmed_authors>Ciblat S</pubmed_authors><pubmed_authors>Clark J</pubmed_authors><pubmed_authors>Chefson A</pubmed_authors><pubmed_authors>Poirier H</pubmed_authors><pubmed_authors>Roulston A</pubmed_authors><pubmed_authors>Morris SJ</pubmed_authors><pubmed_authors>Mochirian P</pubmed_authors><pubmed_authors>Leclaire ME</pubmed_authors><pubmed_authors>Parikh P</pubmed_authors><pubmed_authors>Papp R</pubmed_authors><pubmed_authors>Bubenik M</pubmed_authors><pubmed_authors>Baruah PS</pubmed_authors><pubmed_authors>Patel P</pubmed_authors><pubmed_authors>Gervais C</pubmed_authors><pubmed_authors>Stocco R</pubmed_authors><pubmed_authors>Pinter T</pubmed_authors><pubmed_authors>Sicheri F</pubmed_authors><pubmed_authors>Dorich S</pubmed_authors><pubmed_authors>Gallant M</pubmed_authors><pubmed_authors>Duffy NM</pubmed_authors><pubmed_authors>El Bakkouri M</pubmed_authors><pubmed_authors>Vallee F</pubmed_authors><pubmed_authors>Falgueyret JP</pubmed_authors><pubmed_authors>Barabe F</pubmed_authors><pubmed_authors>Nicolas O</pubmed_authors><pubmed_authors>Mathieu MC</pubmed_authors><pubmed_authors>St-Onge M</pubmed_authors><pubmed_authors>Sfeir A</pubmed_authors><pubmed_authors>Truchon JF</pubmed_authors><pubmed_authors>Pellerin C</pubmed_authors><pubmed_authors>Pau V</pubmed_authors><pubmed_authors>Zahn KE</pubmed_authors><pubmed_authors>Godbout C</pubmed_authors><pubmed_authors>Black WC</pubmed_authors><pubmed_authors>Kurinov I</pubmed_authors><pubmed_authors>Perryman AL</pubmed_authors><pubmed_authors>Mamane Y</pubmed_authors><pubmed_authors>Fortin AB</pubmed_authors><pubmed_authors>Mader P</pubmed_authors><pubmed_authors>Hamel M</pubmed_authors></additional><is_claimable>false</is_claimable>
Identification of <b>RP-6685</b>, an Orally Bioavailable Compound that Inhibits the DNA Polymerase Activity of Polθ.

DNA polymerase theta (Polθ) is an attractive synthetic lethal target for drug discovery, predicted to be efficacious against breast and ovarian cancers harboring BRCA-mutant alleles.
