Unfortunately, current monovalent viral-vectored tuberculosis (TB) vaccine candidates have failed to demonstrate satisfactory clinical protective efficacy. As such, there is a need to develop next-generation viral-vectored TB vaccine strategies which incorporate both vaccine antigen design and delivery route. In this study, we have developed a trivalent chimpanzee adenoviral-vectored vaccine to provide protective immunity against pulmonary TB through targeting antigens linked to the three different growth phases (acute/chronic/dormancy) of Mycobacterium tuberculosis (M.tb) by expressing an acute replication-associated antigen, Ag85</pubmed_abstract><journal>NPJ vaccines</journal><pagination>25</pagination><full_dataset_link>https://www.ebi.ac.uk/biostudies/studies/S-EPMC9948798</full_dataset_link><repository>biostudies-literature</repository><pubmed_title>Intranasal multivalent adenoviral-vectored vaccine protects against replicating and dormant M.tb in conventional and humanized mice.</pubmed_title><pmcid>PMC9948798</pmcid><pubmed_authors>Ottenhoff THM</pubmed_authors><pubmed_authors>Xing Z</pubmed_authors><pubmed_authors>Yang JX</pubmed_authors><pubmed_authors>Chacon A</pubmed_authors><pubmed_authors>Vaseghi-Shanjani M</pubmed_authors><pubmed_authors>Zganiacz A</pubmed_authors><pubmed_authors>Franken KLMC</pubmed_authors><pubmed_authors>D'Agostino MR</pubmed_authors><pubmed_authors>Lepard M</pubmed_authors><pubmed_authors>Jeyanathan M</pubmed_authors><pubmed_authors>Gillgrass A</pubmed_authors><pubmed_authors>Choi MWY</pubmed_authors><pubmed_authors>Afkhami S</pubmed_authors><pubmed_authors>Lai R</pubmed_authors><pubmed_authors>Ertl HC</pubmed_authors></additional><is_claimable>false</is_claimable><name>Intranasal multivalent adenoviral-vectored vaccine protects against replicating and dormant M.tb in conventional and humanized mice.</name><description>Viral-vectored vaccines are highly amenable for respiratory mucosal delivery as a means of inducing much-needed mucosal immunity at the point of pathogen entry.
