How these alterations promote transcriptional programs governing malignancy remains poorly defined. To address this knowledge gap, we evaluated chromatin states across MB subgroups by multi-modal integration of histone modifications with mutational, DNA methylation, and transcriptomic profiles. A bivalent/poised enhancer (EnhBiv) state was specifically enriched at the promoters of neurodevelopmental genes in Groups 3 and 4-MB. Integrative bioinformatics coupled with chromatin occupancy studies identified aberrant KDM2B binding at EnhBiv-enriched promoters. CRISPR-mediated knockout or acute protein degradation of KDM2B selectively suppressed the growth of MB models in vitro and in vivo. Mechanistically, KDM2B promotes sequential recruitment of Polycomb repressive complexes (PRC1/PRC2) and EnhBiv chromatin, thereby repressing neuronal differentiation programs. Collectively, we provide foundational insights into an epigenetic basis of MB, nominating KDM2B as a selective dependency in high-risk subgroups that warrants consideration as a candidate therapeutic target.</description><dates><publication>2026/06/26</publication></dates><accession>GSE270125</accession><cross_references><GSM>GSM9570088</GSM><GSM>GSM9570121</GSM><GSM>GSM9570089</GSM><GSM>GSM9570122</GSM><GSM>GSM9570086</GSM><GSM>GSM9570120</GSM><GSM>GSM9570087</GSM><GSM>GSM9570125</GSM><GSM>GSM9570126</GSM><GSM>GSM9570123</GSM><GSM>GSM9570124</GSM><GSM>GSM9570080</GSM><GSM>GSM9570081</GSM><GSM>GSM9570084</GSM><GSM>GSM9570085</GSM><GSM>GSM9570082</GSM><GSM>GSM9570083</GSM><GSM>GSM9570118</GSM><GSM>GSM9570119</GSM><GSM>GSM9570116</GSM><GSM>GSM9570117</GSM><GSM>GSM9570132</GSM><GSM>GSM9570099</GSM><GSM>GSM9570133</GSM><GSM>GSM9570097</GSM><GSM>GSM9570130</GSM><GSM>GSM9570131</GSM><GSM>GSM9570098</GSM><GSM>GSM9570136</GSM><GSM>GSM9570137</GSM><GSM>GSM9570134</GSM><GSM>GSM9570135</GSM><GSM>GSM9570091</GSM><GSM>GSM8334856</GSM><GSM>GSM8334857</GSM><GSM>GSM9570092</GSM><GSM>GSM8334858</GSM><GSM>GSM8334859</GSM><GSM>GSM9570090</GSM><GSM>GSM9570095</GSM><GSM>GSM9570096</GSM><GSM>GSM9570093</GSM><GSM>GSM9570094</GSM><GSM>GSM9570129</GSM><GSM>GSM9570127</GSM><GSM>GSM9570128</GSM><GSM>GSM9570143</GSM><GSM>GSM8334860</GSM><GSM>GSM9570144</GSM><GSM>GSM8334861</GSM><GSM>GSM9570141</GSM><GSM>GSM8334862</GSM><GSM>GSM9570142</GSM><GSM>GSM8334900</GSM><GSM>GSM8334867</GSM><GSM>GSM8334901</GSM><GSM>GSM8334868</GSM><GSM>GSM8334869</GSM><GSM>GSM8334902</GSM><GSM>GSM8334903</GSM><GSM>GSM8334863</GSM><GSM>GSM9570140</GSM><GSM>GSM8334864</GSM><GSM>GSM8334865</GSM><GSM>GSM8334866</GSM><GSM>GSM9570138</GSM><GSM>GSM9570139</GSM><GSM>GSM8334870</GSM><GSM>GSM8334871</GSM><GSM>GSM8334872</GSM><GSM>GSM8334873</GSM><GSM>GSM8334911</GSM><GSM>GSM8334878</GSM><GSM>GSM8334912</GSM><GSM>GSM8334879</GSM><GSM>GSM8334913</GSM><GSM>GSM8334914</GSM><GSM>GSM8334874</GSM><GSM>GSM8334875</GSM><GSM>GSM8334876</GSM><GSM>GSM8334877</GSM><GSM>GSM8334910</GSM><GSM>GSM8334908</GSM><GSM>GSM8334909</GSM><GSM>GSM8334904</GSM><GSM>GSM8334905</GSM><GSM>GSM8334906</GSM><GSM>GSM8334907</GSM><GSM>GSM8334881</GSM><GSM>GSM8334882</GSM><GSM>GSM8334883</GSM><GSM>GSM8334884</GSM><GSM>GSM9570048</GSM><GSM>GSM9570049</GSM><GSM>GSM8334880</GSM><GSM>GSM8334889</GSM><GSM>GSM8334922</GSM><GSM>GSM8334923</GSM><GSM>GSM8334924</GSM><GSM>GSM8334925</GSM><GSM>GSM8334885</GSM><GSM>GSM8334886</GSM><GSM>GSM8334920</GSM><GSM>GSM8334887</GSM><GSM>GSM8334888</GSM><GSM>GSM8334921</GSM><GSM>GSM8334919</GSM><GSM>GSM8334915</GSM><GSM>GSM8334916</GSM><GSM>GSM8334917</GSM><GSM>GSM8334918</GSM><GSM>GSM8334892</GSM><GSM>GSM9570055</GSM><GSM>GSM8334893</GSM><GSM>GSM9570056</GSM><GSM>GSM8334894</GSM><GSM>GSM9570053</GSM><GSM>GSM9570054</GSM><GSM>GSM8334895</GSM><GSM>GSM9570059</GSM><GSM>GSM9570057</GSM><GSM>GSM8334890</GSM><GSM>GSM8334891</GSM><GSM>GSM9570058</GSM><GSM>GSM9570051</GSM><GSM>GSM8334896</GSM><GSM>GSM8334897</GSM><GSM>GSM9570052</GSM><GSM>GSM8334898</GSM><GSM>GSM9570050</GSM><GSM>GSM8334899</GSM><GSM>GSM8334926</GSM><GSM>GSM8334927</GSM><GSM>GSM9570066</GSM><GSM>GSM9570100</GSM><GSM>GSM9570067</GSM><GSM>GSM9570064</GSM><GSM>GSM9570065</GSM><GSM>GSM9570103</GSM><GSM>GSM9570104</GSM><GSM>GSM9570068</GSM><GSM>GSM9570101</GSM><GSM>GSM9570069</GSM><GSM>GSM9570102</GSM><GSM>GSM9570062</GSM><GSM>GSM9570063</GSM><GSM>GSM9570060</GSM><GSM>GSM9570061</GSM><GSM>GSM9570077</GSM><GSM>GSM9570110</GSM><GSM>GSM9570111</GSM><GSM>GSM9570078</GSM><GSM>GSM9570075</GSM><GSM>GSM9570076</GSM><GSM>GSM9570114</GSM><GSM>GSM9570115</GSM><GSM>GSM9570112</GSM><GSM>GSM9570079</GSM><GSM>GSM9570113</GSM><GSM>GSM9570070</GSM><GSM>GSM9570073</GSM><GSM>GSM9570074</GSM><GSM>GSM9570071</GSM><GSM>GSM9570072</GSM><GSM>GSM9570107</GSM><GSM>GSM9570108</GSM><GSM>GSM9570105</GSM><GSM>GSM9570106</GSM><GSM>GSM9570109</GSM><GPL>24676</GPL><GSE>270125</GSE><taxon>Homo sapiens</taxon></cross_references></HashMap>
