Here, we develop a timestamp tracing method using Rag1CreER knockin mice in combination with Rosa reporters to reveal the processes governing thymic Treg differentiation in native physiological conditions. It enables tracing of thymic T cells at the double negative and double positive stages upon tamoxifen administration. Following labeled cells over time, we define the processes, kinetics, and gene expression changes during thymic T-cell differentiation; specify thymic Treg maturation with CD73, gene expression changes, and Treg-specific DNA demethylation; characterize precursor cells with 4-1BB and/or CD25 expression that are induced by TCR signaling and differentiate to Treg cells in vitro and in vivo; and determine the features of recirculating or retained mature Treg cells. Thus, Rag1CreER–based timestamp tracing delineates thymic Treg ontogeny, proving the concept for exploring T-cell development and heterogeneity with age-resolved precision.</description><dates><publication>2026/07/21</publication></dates><accession>GSE274125</accession><cross_references><GSM>GSM8443923</GSM><GSM>GSM8443924</GSM><GSM>GSM8443921</GSM><GSM>GSM8443932</GSM><GSM>GSM8443933</GSM><GSM>GSM8443922</GSM><GSM>GSM8443930</GSM><GSM>GSM8443931</GSM><GSM>GSM8443920</GSM><GSM>GSM8443918</GSM><GSM>GSM8443929</GSM><GSM>GSM8443919</GSM><GSM>GSM8443916</GSM><GSM>GSM8443927</GSM><GSM>GSM8443928</GSM><GSM>GSM8443917</GSM><GSM>GSM8443925</GSM><GSM>GSM8443914</GSM><GSM>GSM8443926</GSM><GSM>GSM8443915</GSM><GPL>19057</GPL><GSE>274125</GSE><taxon>Mus musculus</taxon><PMID>[42533592]</PMID></cross_references></HashMap>
