To investigate these interactions, we conducted RNA-sequencing of the host cell transcriptome at different time points post-infection. After IAV enters the host cell, it first undergoes transcription (vRNA to mRNA) and then replication (vRNA to cRNA to vRNA). In A549 cells infected with the virus at an MOI of 0.05, RT-PCR assays determined that transcription begins at 3 hours post-infection, the transition from transcription to replication starts at 6 hours, and active replication commences at 12 hours. To elucidate the changes in the host transcriptome during the transcription and replication processes of IAV, we performed RNA-sequencing on cells infected with PR8 virus at 3, 6, and 12 hours post-infection.
