We discovered the epigenetic enzyme Carm1 as a selective negative regulator of cancer antigen presentation by cDC1s, but not cDC2s. Carm1 inactivation promoted cDC1 antigen cross-presentation, activation, and accumulation in tumors; and a Carm1 inhibitor enhanced cDC1-mediated priming of T cells by a cancer vaccine. Carm1 inhibition increased chromatin accessibility at BATF3-JUN and RELA sites critical for cDC1 function and activation. Carm1 expression was regulated by TGF-beta, explaining why Carm1 inactivation enhanced intratumoral cDC1 function without altering cDC1 homeostasis. These studies identify Carm1 as a therapeutic target for enhancing the anti-tumor function of murine and human cDC1.</description><dates><publication>2026/06/30</publication></dates><accession>GSE300014</accession><cross_references><GSM>GSM9052444</GSM><GSM>GSM9052466</GSM><GSM>GSM9052465</GSM><GSM>GSM9052443</GSM><GSM>GSM9052464</GSM><GSM>GSM9052442</GSM><GSM>GSM9052441</GSM><GSM>GSM9052463</GSM><GSM>GSM9052462</GSM><GSM>GSM9052461</GSM><GSM>GSM9052460</GSM><GSM>GSM9052459</GSM><GSM>GSM9052458</GSM><GSM>GSM9052457</GSM><GSM>GSM9052456</GSM><GSM>GSM9052455</GSM><GSM>GSM9052454</GSM><GSM>GSM9052453</GSM><GSM>GSM9052452</GSM><GSM>GSM9052451</GSM><GSM>GSM9052450</GSM><GSM>GSM9052449</GSM><GSM>GSM9052448</GSM><GSM>GSM9052447</GSM><GSM>GSM9052446</GSM><GSM>GSM9052445</GSM><GPL>17021</GPL><GSE>300014</GSE><taxon>Mus musculus</taxon></cross_references></HashMap>
