A thorough understanding of the porcine immune system is needed to safeguard pig health through therapies and vaccines and better utilize swine as preclinical models. Here, we paired single-cell transcriptomic analyses with T and B cell receptor profiling to create a cell atlas of pig hematopoietic development using cells dissociated from central lymphoid organs during embryonic, neonatal, and adult life. We highlight the connection between fetal liver and bone marrow, B cell and gd T cell differentiation trajectories, granulopoiesis in fetal liver, and T and B cell receptor recombination kinetics.
