To understand cooperativity between chemotherapy and immunotherapy, we previously reported a phase II trial sequentially adding pembrolizumab to 5-FU/platinum. In this study, we use single-cell RNA- and TCR-sequencing to analyze 66,813 T cells from primary tumor biopsies pre-treatment, post-chemotherapy, and post-immunotherapy in 33 patients. We observed greater abundance, persistence, and recruitment of T cells with predicted tumor-reactivity in patients with prolonged progression-free survival (slow progressors). Increased B cell abundance and predicted B cell to T cell interactions supported T cell memory and co-stimulation, providing a mechanism for increased abundance and persistence of progenitor-exhausted and tumor-reactive T cells in slow progressors. T cell clones emerging in the tumor after immunotherapy were in the blood before treatment only in slow progressors. Our study thus highlights pre-treatment and early chemotherapy-induced T cell dynamics and B cell to T cell interactions that may drive durable response to chemoimmunotherapy in GC.</description><dates><publication>2026/04/01</publication></dates><accession>GSE315928</accession><cross_references><GSM>GSM9440223</GSM><GSM>GSM9440189</GSM><GSM>GSM9440222</GSM><GSM>GSM9440188</GSM><GSM>GSM9440221</GSM><GSM>GSM9440220</GSM><GSM>GSM9440187</GSM><GSM>GSM9440227</GSM><GSM>GSM9440226</GSM><GSM>GSM9440225</GSM><GSM>GSM9440224</GSM><GSM>GSM9440182</GSM><GSM>GSM9440181</GSM><GSM>GSM9440180</GSM><GSM>GSM9440186</GSM><GSM>GSM9440185</GSM><GSM>GSM9440262</GSM><GSM>GSM9440261</GSM><GSM>GSM9440184</GSM><GSM>GSM9440183</GSM><GSM>GSM9440260</GSM><GSM>GSM9440219</GSM><GSM>GSM9440218</GSM><GSM>GSM9440217</GSM><GSM>GSM9440212</GSM><GSM>GSM9440256</GSM><GSM>GSM9440179</GSM><GSM>GSM9440255</GSM><GSM>GSM9440211</GSM><GSM>GSM9440178</GSM><GSM>GSM9440254</GSM><GSM>GSM9440177</GSM><GSM>GSM9440210</GSM><GSM>GSM9440176</GSM><GSM>GSM9440253</GSM><GSM>GSM9440216</GSM><GSM>GSM9440215</GSM><GSM>GSM9440259</GSM><GSM>GSM9440214</GSM><GSM>GSM9440258</GSM><GSM>GSM9440257</GSM><GSM>GSM9440213</GSM><GSM>GSM9440171</GSM><GSM>GSM9440170</GSM><GSM>GSM9440252</GSM><GSM>GSM9440175</GSM><GSM>GSM9440251</GSM><GSM>GSM9440174</GSM><GSM>GSM9440173</GSM><GSM>GSM9440250</GSM><GSM>GSM9440172</GSM><GSM>GSM9440209</GSM><GSM>GSM9440208</GSM><GSM>GSM9440207</GSM><GSM>GSM9440206</GSM><GSM>GSM9440245</GSM><GSM>GSM9440201</GSM><GSM>GSM9440200</GSM><GSM>GSM9440244</GSM><GSM>GSM9440243</GSM><GSM>GSM9440242</GSM><GSM>GSM9440249</GSM><GSM>GSM9440205</GSM><GSM>GSM9440248</GSM><GSM>GSM9440204</GSM><GSM>GSM9440203</GSM><GSM>GSM9440247</GSM><GSM>GSM9440246</GSM><GSM>GSM9440169</GSM><GSM>GSM9440202</GSM><GSM>GSM9440241</GSM><GSM>GSM9440240</GSM><GSM>GSM9440239</GSM><GSM>GSM9440234</GSM><GSM>GSM9440233</GSM><GSM>GSM9440232</GSM><GSM>GSM9440199</GSM><GSM>GSM9440231</GSM><GSM>GSM9440198</GSM><GSM>GSM9440238</GSM><GSM>GSM9440237</GSM><GSM>GSM9440236</GSM><GSM>GSM9440235</GSM><GSM>GSM9440193</GSM><GSM>GSM9440192</GSM><GSM>GSM9440191</GSM><GSM>GSM9440190</GSM><GSM>GSM9440197</GSM><GSM>GSM9440230</GSM><GSM>GSM9440196</GSM><GSM>GSM9440195</GSM><GSM>GSM9440194</GSM><GSM>GSM9440229</GSM><GSM>GSM9440228</GSM><GPL>24676</GPL><GSE>315928</GSE><taxon>Homo sapiens</taxon></cross_references></HashMap>
