The catalytic activity of TET1 is required for complete demethylation in PGCs, yet sperm from Tet1-/- mice display methylation defects not explained by incomplete erasure. Instead, these defects arise from abnormal de novo methylation during development, coinciding with erosion of H3K4me3, a chromatin modification that blocks DNMT3A/3L. Using a catalytically inactive Tet1HxD mouse line, we demonstrate a non-catalytic role of TET1 in promoting H3K4me3 deposition and protecting a subset of sperm hypomethylated regions from aberrant methylation in prospermatogonia.","dates":{"publication":"2026/07/22"},"accession":"GSE318253","cross_references":{"GSM":["GSM9491413","GSM9491402","GSM9491403","GSM9491414","GSM9491411","GSM9491400","GSM9491401","GSM9491412","GSM9491406","GSM9491407","GSM9491404","GSM9491415","GSM9491405","GSM9491416","GSM9491408","GSM9491409","GSM9491399","GSM9491410"],"GPL":["32159"],"GSE":["318253"],"taxon":["Mus musculus"],"PMID":["[42448565]"]}}
