The catalytic activity of TET1 is required for complete demethylation in PGCs, yet sperm from Tet1-/- mice display methylation defects not explained by incomplete erasure. Instead, these defects arise from abnormal de novo methylation during development, coinciding with erosion of H3K4me3, a chromatin modification that blocks DNMT3A/3L. Using a catalytically inactive Tet1HxD mouse line, we demonstrate a non-catalytic role of TET1 in promoting H3K4me3 deposition and protecting a subset of sperm hypomethylated regions from aberrant methylation in prospermatogonia.","dates":{"publication":"2026/07/22"},"accession":"GSE319129","cross_references":{"GSM":["GSM9511531","GSM9511532","GSM9511540","GSM9511530","GSM9511539","GSM9511528","GSM9511529","GSM9511526","GSM9511537","GSM9511527","GSM9511524","GSM9511535","GSM9511536","GSM9511533","GSM9511523","GSM9511534"],"GPL":["31191"],"GSE":["319129"],"taxon":["Mus musculus"],"PMID":["[42448565]"]}}
