Rothia mucilaginosa, a commensal bacterium with established anti-inflammatory properties, has not been previously investigated for its effects on intestinal function. In this study, we evaluated the therapeutic potential of R. mucilaginosa in a loperamide-induced constipation mouse model using multiomics approaches. Twenty-six SPF male C57BL/6 mice were divided into normal control (NC, n=8), constipation model control (MC, n=8), and R. mucilaginosa-treated (RG, n=10) groups. R. mucilaginosa intervention significantly improved fecal output and induced gut microbiota remodeling, including enrichment of Akkermansia muciniphila and Alistipes finegoldii. To characterize host molecular responses, RNA-seq was performed on colon tissues to identify differentially expressed genes and pathways associated with constipation alleviation, with particular focus on neuroactive pathway activation.</description><dates><publication>2026/04/27</publication></dates><accession>GSE319612</accession><cross_references><GSM>GSM9521330</GSM><GSM>GSM9521352</GSM><GSM>GSM9521353</GSM><GSM>GSM9521331</GSM><GSM>GSM9521332</GSM><GSM>GSM9521354</GSM><GSM>GSM9521333</GSM><GSM>GSM9521355</GSM><GSM>GSM9521350</GSM><GSM>GSM9521351</GSM><GSM>GSM9521338</GSM><GSM>GSM9521339</GSM><GSM>GSM9521334</GSM><GSM>GSM9521335</GSM><GSM>GSM9521336</GSM><GSM>GSM9521337</GSM><GSM>GSM9521341</GSM><GSM>GSM9521342</GSM><GSM>GSM9521343</GSM><GSM>GSM9521344</GSM><GSM>GSM9521340</GSM><GSM>GSM9521349</GSM><GSM>GSM9521345</GSM><GSM>GSM9521346</GSM><GSM>GSM9521347</GSM><GSM>GSM9521348</GSM><GPL>24247</GPL><GSE>319612</GSE><taxon>Mus musculus</taxon></cross_references></HashMap>
