Human M1- or M2-polarized macrophages were co-cultured with RA fibroblast-like synoviocytes (RAFLS) and endothelial cells (HUVECs) to generate spheroids embedded in a collagen-based matrix (n=4). Spheroids were exposed to inflammatory stimuli (20%SF) and pharmacological inhibitors including JAK1/3 inhibitor tofacitinib (5μM) and NF-ĸB pathway inhibitors IKKβi (2.5μM) and NIKi (5μM).
