We have previously shown that honokiol upregulates mitochondrial sirtuin 3 (SIRT3) and protects against CIHL. In the present study, we further investigated the mechanisms of honokiol and the role of Sirt3 in protection against CIHL by using a Sirt3 knockout (Sirt3−/−) mouse model and by performing RNA-seq on outer hair cells. Our results showed that honokiol ameliorated cisplatin-induced cochlear functional deterioration in both wild-type and Sirt3−/− mice. RNA-seq analysis showed that cisplatin treatment resulted in the downregulation of mitochondrial-related transcriptome and important antioxidant genes, such as isocitrate dehydrogenase 2 (Idh2). Honokiol pre-treatment reversed this pattern by upregulating genes suppressed by cisplatin. These results highlight the clinical potential of honokiol as a protective agent during chemotherapy, while suggesting that Idh2, a substrate of Sirt3, is a promising therapeutic target.</description><dates><publication>2026/08/19</publication></dates><accession>GSE328003</accession><cross_references><GSM>GSM9670209</GSM><GSM>GSM9670208</GSM><GSM>GSM9670229</GSM><GSM>GSM9670228</GSM><GSM>GSM9670227</GSM><GSM>GSM9670237</GSM><GSM>GSM9670215</GSM><GSM>GSM9670214</GSM><GSM>GSM9670236</GSM><GSM>GSM9670235</GSM><GSM>GSM9670213</GSM><GSM>GSM9670234</GSM><GSM>GSM9670212</GSM><GSM>GSM9670211</GSM><GSM>GSM9670233</GSM><GSM>GSM9670232</GSM><GSM>GSM9670210</GSM><GSM>GSM9670231</GSM><GSM>GSM9670230</GSM><GSM>GSM9670219</GSM><GSM>GSM9670218</GSM><GSM>GSM9670239</GSM><GSM>GSM9670217</GSM><GSM>GSM9670216</GSM><GSM>GSM9670238</GSM><GSM>GSM9670226</GSM><GSM>GSM9670225</GSM><GSM>GSM9670224</GSM><GSM>GSM9670223</GSM><GSM>GSM9670222</GSM><GSM>GSM9670221</GSM><GSM>GSM9670220</GSM><GPL>34290</GPL><GSE>328003</GSE><taxon>Mus musculus</taxon></cross_references></HashMap>
