Here, we identify the histone acetyltransferase RTT109 as a key regulator of PRC2-repressed domains in the model fungus Neurospora crassa. Although RTT109 interacts with the VPS75 homolog Nucleosome Assembly Factor 2 (NAF-2), we show that proper structure and function of PRC2-methylated chromatin require RTT109 catalytic activity but are independent of NAF-2 and H3K56 acetylation. We further demonstrate that H3K27me3 can be stably propagated over multiple rounds of mitosis in the absence of sequence-specific PRC2 targeting, and that RTT109 is essential for maintenance of the repressed state. These findings uncover a replication-linked mechanism for epigenetic memory and establish RTT109 as a key regulator of Polycomb-mediated chromatin inheritance.</description><dates><publication>2026/05/17</publication></dates><accession>GSE330894</accession><cross_references><GSM>GSM9735250</GSM><GSM>GSM9735142</GSM><GSM>GSM9735143</GSM><GSM>GSM9735141</GSM><GSM>GSM9735148</GSM><GSM>GSM9735149</GSM><GSM>GSM9735146</GSM><GSM>GSM9735147</GSM><GSM>GSM9735144</GSM><GSM>GSM9735145</GSM><GSM>GSM9735230</GSM><GSM>GSM9735197</GSM><GSM>GSM9735231</GSM><GSM>GSM9735198</GSM><GSM>GSM9735195</GSM><GSM>GSM9735196</GSM><GSM>GSM9735193</GSM><GSM>GSM9735194</GSM><GSM>GSM9735191</GSM><GSM>GSM9735192</GSM><GSM>GSM9735190</GSM><GSM>GSM9735238</GSM><GSM>GSM9735239</GSM><GSM>GSM9735236</GSM><GSM>GSM9735237</GSM><GSM>GSM9735234</GSM><GSM>GSM9735235</GSM><GSM>GSM9735199</GSM><GSM>GSM9735232</GSM><GSM>GSM9735233</GSM><GSM>GSM9735241</GSM><GSM>GSM9735242</GSM><GSM>GSM9735240</GSM><GSM>GSM9735249</GSM><GSM>GSM9735247</GSM><GSM>GSM9735248</GSM><GSM>GSM9735245</GSM><GSM>GSM9735246</GSM><GSM>GSM9735243</GSM><GSM>GSM9735244</GSM><GSM>GSM9735175</GSM><GSM>GSM9735176</GSM><GSM>GSM9735173</GSM><GSM>GSM9735174</GSM><GSM>GSM9735171</GSM><GSM>GSM9735172</GSM><GSM>GSM9735170</GSM><GSM>GSM9735218</GSM><GSM>GSM9735219</GSM><GSM>GSM9735216</GSM><GSM>GSM9735217</GSM><GSM>GSM9735214</GSM><GSM>GSM9735215</GSM><GSM>GSM9735179</GSM><GSM>GSM9735212</GSM><GSM>GSM9735213</GSM><GSM>GSM9735210</GSM><GSM>GSM9735177</GSM><GSM>GSM9735211</GSM><GSM>GSM9735178</GSM><GSM>GSM9735186</GSM><GSM>GSM9735187</GSM><GSM>GSM9735220</GSM><GSM>GSM9735184</GSM><GSM>GSM9735185</GSM><GSM>GSM9735182</GSM><GSM>GSM9735183</GSM><GSM>GSM9735180</GSM><GSM>GSM9735181</GSM><GSM>GSM9735229</GSM><GSM>GSM9735227</GSM><GSM>GSM9735228</GSM><GSM>GSM9735225</GSM><GSM>GSM9735226</GSM><GSM>GSM9735223</GSM><GSM>GSM9735224</GSM><GSM>GSM9735188</GSM><GSM>GSM9735221</GSM><GSM>GSM9735222</GSM><GSM>GSM9735189</GSM><GSM>GSM9735153</GSM><GSM>GSM9735154</GSM><GSM>GSM9735151</GSM><GSM>GSM9735152</GSM><GSM>GSM9735150</GSM><GSM>GSM9735159</GSM><GSM>GSM9735157</GSM><GSM>GSM9735158</GSM><GSM>GSM9735155</GSM><GSM>GSM9735156</GSM><GSM>GSM9735164</GSM><GSM>GSM9735165</GSM><GSM>GSM9735162</GSM><GSM>GSM9735163</GSM><GSM>GSM9735160</GSM><GSM>GSM9735161</GSM><GSM>GSM9735209</GSM><GSM>GSM9735207</GSM><GSM>GSM9735208</GSM><GSM>GSM9735205</GSM><GSM>GSM9735206</GSM><GSM>GSM9735203</GSM><GSM>GSM9735204</GSM><GSM>GSM9735168</GSM><GSM>GSM9735201</GSM><GSM>GSM9735202</GSM><GSM>GSM9735169</GSM><GSM>GSM9735166</GSM><GSM>GSM9735167</GSM><GSM>GSM9735200</GSM><GPL>35294</GPL><GSE>330894</GSE><taxon>Neurospora crassa</taxon></cross_references></HashMap>
