Patient-derived human intestinal organoids from four independent donors were differentiated into colonocyte-enriched organoids and engineered to overexpress BCL2, thereby limiting apoptosis-associated cell death. Organoids were then exposed to IFNγ and TNF to model an inflammatory cytokine environment relevant to intestinal inflammation, in the presence or absence of pharmacological inhibitors targeting necroptotic signalling. Transcriptomic profiling was performed to determine how inhibition of necroptosis-associated pathways alters epithelial inflammatory responses under conditions where apoptosis is blocked.
