Blood samples were drawn within 6 days of delivery (t0) and at 12 weeks postpartum (t4). Participants were assessed longitudinally for postnatal depression (EPDS), perceived stress (PSS-10), and mother-infant attachment (MPAS). A subset of N = 247 participants with complete blood sampling was used for methylation analysis; mothers with the most severe depressive symptoms (PPD group, n = 17, DSM-5 confirmed) were compared to a euthymic healthy control group (HC, n = 24). Genome-wide methylation profiling was performed using the Illumina Infinium Human MethylationEPICv2 BeadChip. Differential methylation analysis was conducted using the ChAMP package (BMIQ normalization, limma-based DMP calling).
