The pathway consists of multigene families of trehalose phosphate synthases (TPSs) and trehalose phosphate phosphatases (TPPs). TPSs consist of class I and II enzymes. Class I enzymes are catalytically active but the function of class II TPSs has been enigmatic as they do not possess catalytic activity. To decipher physiological function of class II TPSs in cereals, we target a clade of three TPSs - OsTPS2, OsTPS3 and OsTPS6 in rice - with closest similarity to Arabidopsis TPS7. Gene editing with CRISPR-cas9 of OsTPS2, OsTPS3 and OsTPS6 together resulted in delayed flowering and poor grain set. Investigation of pollen showed abnormal structure, poor adherence to stigmas and reduced starch accumulation. RNAseq analysis of panicles at pre-anthesis developmental stages showed large effects on genes involved in flowering time, pollen viability and reproductive development.
