To investigate the molecular mechanisms governing H3mm15 deposition and genomic localization, we performed Chromatin Integration Labeling followed by sequencing (ChIL-seq) in NIH3T3 cells expressing EGFP-tagged H3.3, H3mm15, or H3mm15 single-reversion mutants. The dataset consists of two complementary experiments. First, H3.3-EGFP- and H3mm15-EGFP-expressing cells were treated with siRNAs targeting the histone chaperones Daxx or Hira and compared with cells treated with a non-targeting control siRNA. The genomic incorporation of the EGFP-tagged histones was profiled by GFP-ChIL-seq. Second, the genomic distributions of H3mm15-EGFP and three single-reversion mutants, Q100L, V112I, and C128R, were compared with that of H3.3-EGFP.
