Subunit Med15 is required for pre-natal development and post-natal maturation of pancreatic β-cells. However, whether Med15 continues to play a role in β-cell function after maturation is complete, and throughout adulthood, is unknown. To investigate the role of Med15 in β-cells post-maturation, we induced a β-cell specific Med15 knockout at six weeks age in male and female mice. Ablation of Med15 led to glucose intolerance and impaired insulin secretion. RNA-sequencing revealed downregulation of β-cell maturation markers. Further, we implanted insulin pellets into knockout mice to lower blood glucose and performed RNA-seq to validate that the transcriptional changes were a direct consequence of the Med15 loss and not due to hyperglycemia in the knockout mice. This study shows that Med15 is required after weaning and throughout life to maintain functional β-cell maturity.</description><dates><publication>2026/08/19</publication></dates><accession>GSE343956</accession><cross_references><GSM>GSM9965560</GSM><GSM>GSM9965563</GSM><GSM>GSM9965564</GSM><GSM>GSM9965561</GSM><GSM>GSM9965562</GSM><GSM>GSM9965545</GSM><GSM>GSM9965567</GSM><GSM>GSM9965568</GSM><GSM>GSM9965546</GSM><GSM>GSM9965565</GSM><GSM>GSM9965566</GSM><GSM>GSM9965549</GSM><GSM>GSM9965569</GSM><GSM>GSM9965547</GSM><GSM>GSM9965548</GSM><GSM>GSM9965570</GSM><GSM>GSM9965571</GSM><GSM>GSM9965552</GSM><GSM>GSM9965553</GSM><GSM>GSM9965572</GSM><GSM>GSM9965550</GSM><GSM>GSM9965551</GSM><GSM>GSM9965556</GSM><GSM>GSM9965557</GSM><GSM>GSM9965554</GSM><GSM>GSM9965555</GSM><GSM>GSM9965558</GSM><GSM>GSM9965559</GSM><GPL>30172</GPL><GSE>343956</GSE><taxon>Mus musculus</taxon></cross_references></HashMap>
