Control (young) and replicatively senescent endothelial cells were co-cultured with human macrophages in endothelial cell–macrophage spheroids. Single-cell transcriptomic profiling was used to identify major endothelial and macrophage populations and to characterize cell-state-specific gene expression patterns. Cell–cell communication analysis was subsequently performed using CellChat to investigate potential intercellular signaling networks within the co-culture system.
