Here, we identify a serine/threonine protein phosphatase catalytic subunit, Pp4c, as essential for growth, conidiation, virulence, and the utilization of carbohydrates and lipids in Magnaporthe oryzae. We demonstrate that the protein phosphatase 4 complex (Pp4c and Smek1 subunits), the AMP-activated protein kinase (AMPK) Snf1, and the transcriptional regulators CreA (repressor) and Crf1 (activator) collaboratively regulate the utilization of non-preferred carbon sources. Protein interaction and phosphorylation analyses reveal that under glucose-rich conditions, Snf1 and Smek1 directly regulate the phosphorylation status of CreA and Crf1. In contrast, under L-arabinose-rich conditions, Snf1 indirectly modulates the dephosphorylation of these transcription factors via Pp4c and Smek1. Phosphorylation-mediated activation or inactivation of CreA and Crf1 drives CCR and CCDR, thereby governing the metabolism of carbon sources derived from plant cell walls and contributing to fungal pathogenicity. These findings provide deep insights into the regulation of CCR and CCDR, emphasizing their significance in carbon metabolism and pathogenicity in phytopathogenic fungi."],"pubmed_title":["A multilayered regulatory network mediated by protein phosphatase 4 controls carbon catabolite repression and de-repression in Magnaporthe oryzae."],"pubmed_authors":["Huang Zhicheng Z, Wang Qing Q, Li Yan Y, Huang Pengyun P, Liao Jian J, Wang Jing J, Li Hui H, Cai Yingying Y, Wang Jiaoyu J, Liu Xiaohong X, Lin Fu-Cheng FC, Lu Jianping J"],"name_synonyms":["CRFR1, Mass Spectrum Analysis, Peptidomics., Mass Spectrum, CRF1, CRH-R1, Analyses, CRF-R1, CRHR1L, Spectrometry, cytokinin response factor 1, Spectrum Analyses, CRH-R-1, Spectrum Analysis, Spectroscopy, CRFR-1, CRHR, MS, CRF-R, CRH-R1h, Mass, CRHR1f, Analysis, T22B4_120, Mass Spectrum Analyses, Mass Spectroscopy, T22B4.120, CRF-R-1"],"pubmed_abstract_synonyms":["3.1.3.16, biochemical pathways, Cognitive Function, Metabolic Process, Memory, STPP, protein kinase A activity, single-organism developmental process, 5'-AMP-Activated Protein Kinase, beta-hydroxy-beta-methylglutaryl-CoA reductase kinase activity, Glukose, postnatal development, Metabolic Concepts, crotonyl-coenzyme A reductase activity, Monohydrate, Pp4, AMPKa2, growth and development, Carbon Catabolite Repression, protein, CG32505, (DL)-Isomer, AMPKa1, ATP:hydroxymethylglutaryl-CoA reductase (NADPH) phosphotransferase activity, infectivity, phosphorylation, Social Controls, I-peptide kinase activity, Magnaporthe oryzae, Regions, Carbon-12, cAMP-dependent protein kinase, EMB2736, CRH-R1h, Dextrose, CG9063, PP1, symptoms, Concepts, smk-1, PP4, pp4, AMP-Activated Protein Kinase alpha Subunit, AMP-Activated Protein Kinase, Metabolism Concept, protein aggregate, PpX, T22B4_120, Phenomenon, Protein phosphatase X, Formal Social Controls, Catalytic Core, CG1459, Cell Walls, viral infection, C, HAMPKb, CG3051, CRH-R1, Repression, Subunit, catabolism, PP4 19C, ATP:acetyl-CoA carboxylase phosphotransferase activity, Catabolite, NCPR, AMPK2, 3', Subunits, metabolic process resulting in cell growth, CG18339, 1110034C04Rik, acetyl-CoA carboxylase bound kinase activity, PPX, FLFL1, BcDNA:GH03694, DL-glucose, CCR1, Social, ampk2, glucose, False Memory, Cognitions, 5' cAMP-dependent protein kinase activity, Ampka2, dAMPKa, CRF-R, Functions, Carbon, AMP Activated Protein Kinase beta Subunits, 5' AMP Activated Protein Kinase, L-Arabinose, Delayed Memory, biotransformation, PPP4c, snf1a, Catalytic Subunits, Catabolism, HMG-CoA reductase kinase activity, DmelCG9063, CRF-R-1, screening, 3-hydroxy-3-methylglutaryl-CoA reductase kinase activity, Molds, carbon catabolite repression, Process, statistics and numerical data, PP4R3A, BC064465, metabolism resulting in cell growth, Fungi, DmelCG11567, DmPpp4-19C, AMP-Activated Protein Kinase beta Subunit, dephosphorylation, Insight, Memories, Mold, Vitreous, PAS14, Repressed, CRFR-1, Catalytic Regions, PP4C, Repressed Memory, Lipid, Pathogenicity, Domains, AMP Activated Protein Kinase, numerical data, secretion, (alpha-D)-Isomer, Region, acetyl-CoA carboxylase kinase (AMP-activated) activity, Domain, activation, AMP-Activated Protein Kinase gamma Subunit, EMBRYO DEFECTIVE 2736, AMP Activated Kinase, D-Glucose, PKA C, Mold., Factors, Drives, growth pattern, EG:132E8.2, non-developmental growth, 6C, PPH3, BcDNAGH03694, Catabolite Repression, Control, Reactive Site, signs, prkaa, acetyl-CoA carboxylase kinase-3 (AMP-activated) activity, 2002, Controls, L Arabinose, 3L6, DmelCG3051, Delayed, PRKAA, Vitreous Carbon, ATP:protein phosphotransferase (cAMP-dependent) activity, DMR, Filamentous, rich, D Glucose, AMP-Activated Protein Kinase alpha Subunits, AMP Activated Protein Kinase alpha Subunit, ampka1, KIAA2010, carbonium, acetyl-CoA carboxylase kinase 2 activity, CG1596, dAMPKalpha, ACK3, acetyl-CoA carboxylase kinase activity, Reactive Sites, ACK2, Glucose Monohydrate, Regulation, adenosine 3', gluco-hexose, Regulations, Magnaporthe oryzae B.C.
