CatSper comprises the pore-forming proteins CATSPER1-4 and multiple auxiliary subunits, including CATSPERβ, γ, δ, ε, ζ, and EFCAB91,3-9. Here we report the cryo-electron microscopy (cryo-EM) structure of the CatSper complex isolated from mouse sperm. In the extracellular view, CATSPER1-4 conform to the conventional domain-swapped voltage-gated ion channel fold10, following a counterclockwise arrangement. The auxiliary subunits CATSPERβ, γ, δ and ε-each of which contains a single transmembrane segment and a large extracellular domain-constitute a pavilion-like structure that stabilizes the entire complex through interactions with CATSPER4, 1, 3 and 2, respectively. Our EM map reveals several previously uncharacterized components, exemplified by the organic anion transporter SLCO6C1. We name this channel-transporter ultracomplex the CatSpermasome. The assembly and organization details of the CatSpermasome presented here lay the foundation for the development of CatSpermasome-related treatments for male infertility and non-hormonal contraceptives."],"pubmed_title":["Structure of a mammalian sperm cation channel complex."],"pubmed_authors":["Lin Shiyi S, Ke Meng M, Zhang Yuqi Y, Yan Zhen Z, Wu Jianping J"],"pubmed_abstract_synonyms":["Cryomicroscopies, Kation, Male, methionine aminopeptidase activity, Laboratory, Ass-1, FON1, cation, Mus domesticus, postnatal development, Mbp1, Non-Governmental Organizations, Organic Anion Transport Polypeptide, growth and development, Progress Reports, FLORAL ORGAN NUMBER 1, Foundation, House Mouse, World Fertility Survey, Fertility Preferences, serial element, Below Replacement, Summary Report, Ionic Channels, AA408052, SUP, fold, 2, Infertility, myd, Effect, Summary Reports, Fertility Surveys, peptidase M activity, Transporter, male, Transport Proteins, Male Infertility, X-Chromosome-Bearing Sperm, adenomas, sperm cell, Fertility Incentives, rabGAPLP, X-Bearing, L-methionine aminopeptidase activity, pore, Progress Report, Natural, membrane region, Bdr, CatSper channel complex, Mbp-1, RabGAP-5, Swiss Mice, Super protein, sp, Sub-Fertility, SNAP-25, ASS, Progress, Y Chromosome Bearing Sperm, Field Reports, \"male infertility\" EXACT [SNOMEDCT_2005_07_31:155924001], Y-Bearing Sperm, X Chromosome Bearing Sperm, RUSC3, infertility, Y-Chromosome-Bearing Sperms, Ionic Channel, Organic Anion Transport Protein, Fertility Determinant, Therapies, Fertility Preference, CatSper1, Cryoelectron, OATP Proteins, house mouse, Membrane Transport Proteins, Organizations, Differential Fertility, ejaculate, Therapy, Y-Chromosome-Bearing, Motility, sterility, OATP Transporters, organisation, gyltl1b-b, World Fertility Surveys, mouse, MYH-associated polyposis, autosomal recessive familial adenomatous polyposis, Contraceptives, Cryo-electron, integral to membrane, Maps, OAT Transport Protein, segment, Organic Anion, male reproductive system infertility disorder, Investigative Report, Cryomicroscopy, MDDGA6, mKIAA0609, Electron Cryomicroscopy, GENA70, Cryoelectron Microscopies, Anion Transporter, KIAA0609, autosomal recessive, fg, Organic Anion Transport Proteins, Mus musculus, X-Chromosome-Bearing, RUTBC3, OAT, gyltl1b, sperm movement, Nongovernmental, mice, Channels, administrative structure, Field, Swiss Mouse, mdc1d, expanded, Membrane Channels, Electron, infertility disorder of male reproductive system, spermatozoid, X Bearing Sperm, Preferences, Treatments, Fertility Determinants, domesticus, RABGAP5, seminal fluid, MDC1D, Report, enr, \"Infertility, Agents, enlarged, unspecified\" EXACT [ICD9CM_2006:606.9], Sperms, segment -
