Despite their ecological importance, detailed proteomic characterization on diatoms has remained limited. Here, we report a method for the isolation of mitochondrial complex I from P. tricornutum and present a comprehensive proteomic analysis. We create two GelMaps allowing identification of protein complex subunits from a organelle fraction as well as from a complex I enriched fraction. Our results confirm the presence of a carbonic anhydrase (CA) module, previously proposed as an ancestral feature of complex I, and identify one novel, clade-specific subunit that may contribute additional functionality.</description><dates><publication>Wed Aug 27 06:08:00 BST 2025</publication></dates><accession>MSV000098949</accession><cross_references/></HashMap>
