The column was set at 40°C, and the injection volume was 2 μL. The eluents for the positive and negative polarity mode were 0.1% formic acid in water (v/v, A2) and 0.1% formic acid in acetonitrile (v/v, B2). Three cycles of treatment were performed: freezing in liquid nitrogen for 5 min, followed by thawing at room temperature and grinding at 55 Hz for 60 s. The mixture was then centrifuged at 12,000 rpm and 4°C for 15 min. Subsequently, 800 μL of the supernatant to dryness under vacuum, and reconstitute with 150 μL of 50% methanol containing 5 ppm 2-chlorophenylalanine by vortexing for 30 s.
