The mixture was vortexed for 30 s, homogenized at 35 Hz for 4 min, and sonicated for 5 min in an ice-water bath. The homogenization and sonication steps were repeated three times. Samples were then incubated at -40 °C for 1 h and centrifuged at 12,000 rpm, 4 °C, for 15 min. The supernatant was transferred to sample vials for LC-MS/MS analysis. Immediately following collection, the fresh specimens were flash-frozen in liquid nitrogen to preserve metabolic profiles and stored at -80°C in an ultra-low temperature freezer, with no freeze-thaw cycles prior to non-targeted metabolomic analysis.
