Tumor-derived macrophage/microglia were isolated as mentioned previously. Briefly, samples were initially dissociated into fragments under sterile conditions using scissors. Then, tissue fragments were digested to be suspensions using papain. The suspensions were resuspended in cold sterile PBS and filtered through a 40-μm strainer cap, followed by centrifugation for 5 min at 300 g. CD11b+ cells were sorted using CD11b microbeads (Miltenyi Biotec, 130-093-634, Germany) followed by procedures provided by the manufacturer. Finally, the total RNA of CD11b+ cells were extracted and used for RNA sequencing at illumina platform.
