The process includes single cell encapsulation, mRNA capture, reverse transcription, and cDNA amplification in droplet emulsion. Libraries for each sample were indexed and sequenced using a MGISEQ-2000 sequencer (MGI). Raw reads embedded in fastq files were preprocessed using the pipeline of DNBelab C Series HT singlecell analysis software (https://github.com/MGI-tech-bioinformatics/DNBelab_C_Series_HT_scRNA-analysis-software, V1.2). The mouse reference genome GRCm38 and gene annotation (Release M23) downloaded from GENCODE database were used to align the reads and quantify the gene expression.
