Conflicting data on age-related changes in circulating levels have been reported mainly due to the lack of specific detection methods. More recently, liquid chromatography tandem mass spectrometry (LC-MS/MS) based assay showed that the circulating levels of GDF11 do not change significantly throughout human lifespan, but GDF8 levels decrease with aging in men. Here a novel detection method is demonstrated based on parallel reaction monitoring LC-MS/MS assay combined with immunoprecipitation to reliably distinguish GDF11 and GDF8 as well as determine their endogenous levels in mouse serum. The data indicate that both GDF11 and GDF8 circulating levels significantly decline with aging in female mice.</pubmed_abstract><pubmed_title>Targeted Approach to Distinguish and Determine Absolute serum</name><description>Abstract
Growth differentiation factor 11 (GDF11) and Myostatin (GDF8) are circulating factor shown to reduce cardiac hypertrophy of aged animals. Although GDF11 and GDF8 have high structural homology, it seems that GDF11 is had a more potent effect on reducing cardiomyocytes size, albeit GDF11 quantification methods have been recently questioned. In this study, we developed the IP based-PRM- LC-MS/MS method for measuring the GDF8 and GDF11 in one assay. The method demonstrated discriminatory power and allow the quantification of both proteins. Comparison of young and old group of mice show declination of both, GDF8 and GDF11 during aging.</description><dates><publication>Sun Jan 24 00:00:00 GMT 2021</publication></dates><accession>PXD015838</accession><cross_references><TAXONOMY>10090</TAXONOMY><pubmed>32104967</pubmed></cross_references></HashMap>
