Initially, the worms were collected in three biological replicas per genotype and processed in order to extract their proteins at a whole proteome level by subjecting the individuals to homogenisation and complete lysis in 4%SDS and 100mM DTT assisted by a heating (99oC; 5min) and sonication. Homogenates were centrifuged at 17000x g for 15 minutes and the supernatants/protein extracts collected and processed according to the Sp3 protocol. The Sp3 protocol result in the tryptic digestion of the proteins, that after a Sp3 mediated peptide cleanup are ready to be analysed through LC-MS/MS operating in DIA mode. Our system consists of a Dionex Ultimate 3000RSLC online with a Thermo Q Exactive HF-X Orbitrap mass spectrometer. The pure peptidic samples were directly injected and separated on an 25 cm-long analytical C18 column (PepSep, 1.9μm3 beads, 75  µm ID) using an one-hour long run, starting with a gradient  of 7% Buffer B (0.1% Formic acid in 80% Acetonitrile) to 35% for 40 min and followed by an increase to 45% in 5 min and a second increase to 99% in 0.5min and then kept constant for equilibration for 14.5min. A full MS was acquired in profile mode using a Q Exactive HF-X Hybrid Quadropole-Orbitrap mass spectrometer, operating in the scan range of 375-1400 m/z using 120K resolving power with an AGC of 3x 106 and max IT of 60ms followed by data independent analysis using 8 Th windows (39 loop counts) with 15K resolving power with an AGC of 3x 105 and max IT of 22ms and a normalized collision energy (NCE) of 26."],"repository":["Pride"],"quantification_method":["Not available"],"modification":[""],"data_protocol":["Orbitrap raw data was analysed in DIA-NN 1.9.2 (Data-Independent Acquisition by Neural Networks) (34) through searching against the reference C. elegans and E. coli  Uniprot databases supplemented with the common contaminant database. The search was performed using the library-free mode of the software, allowing up to two tryptic missed cleavages. A spectral library was created by the DIA runs and used to reanalyze them. DIA-NN default settings have been used with oxidation of methionine residues and acetylation of the protein N-termini set as variable modifications and carbamidomethylation of cysteine residues as fixed modification. N-terminal methionine excision was also enabled. The match between runs (MBR) feature was used for all analyses and the output (precursor) was filtered at 1% FDR and finally the protein inference was performed on the level of genes using only proteotypic (unique) peptides. While pathological Tau mutations are known to impair mitochondrial dynamics and function, the physiological role of wild-type Tau in the maintenance of mitochondrial homeostasis remains poorly understood. Here, using C. elegans and mice lacking PTL-1, the nematode Tau-like homolog, and Tau respectively, we demonstrate that Tau deficiency enhances mitochondrial respiration, ATP production and mitophagy, promoting a pro-fusion mitochondrial network in neurons. Tau-deficient nematodes also display increased resistance to stressors, including heat and mitochondrial insults. Strikingly, loss of FZO-1, the mitofusin homolog, abolishes these effects, reducing mitochondrial function and stress resistance in PTL-1 deficient nematodes. Our findings reveal a conserved role for wild-type Tau in restraining mitochondrial fusion and activity via mitofusins, highlighting its contribution to mitochondrial quality control and cellular stress
