Here, we generate mice in which TCF-1 fails to become downregulated in differentiated effector cells and probe the transcriptional impact of TCF-1 retention in different T cell differentiation states by scRNA-sequencing. Overall design: CD8+ T cells specific to the LCMV-gp3 antigen were sorted from the spleens of two WT control and two Tcf7∆+22kb mice on 8 dpi with LCMV-Armstrong.
