Despite clinical manifestations in patients, traditional diagnostic methods failed to confirm the infection due to the absence of S. japonicum eggs in fecal samples and the lack of the intermediate host Oncomelania hupensis locally. The research examined rectal biopsy samples stored for 6-16 years, which were severely degraded and unsuitable for conventional sequencing.
