We explored the expression and functional aspects of miR-200c in genesis of leiomyomas, benign uterine tumors with fibrotic characteristic. Gain-of function of miR-200c in isolated leiomyoma and myometrial smooth muscle cells (LSMC and MSMC) and leiomyosarcoma cell line (SKLM-S1) repressed ZEB1/ZEB2 mRNAs and proteins, with concurrent increase in E-cadherin (CDH1) and reduction in vimentin expression, phenotypic alteration and inhibition of MSMC and LSMC proliferation. We further validated TIMP2, FBLN5 and VEGFA as direct targets of miR-200c through interaction with their respective 3’UTRs, and other genes as determined by microarray analysis. Overall design: Total RNA isolated from MSMC and LSMC following gain-of function of miR-200c was subjected to gene expression profiling using HumanHT-12 v4 Expression BeadChip (Illumina, Inc. San Diego, CA) consists of 47,000 oligonucleotide probe sets representing 28,688 transcripts.</long_description><tag>xref:PubMed:22685266</tag><repository>ENA</repository><description_synonyms>9430057C20Rik, HH, TIMP-2, l(3)hh, ARCL1A, Vegf164, Vegf120, Myosin regulatory light chain interacting protein, EVEC., 6.3.2.-, ARMD3, bar, TIMP2, IDOL, CG4637, Hh, CG12249, A55, AW228700, FIBL-5, tissue inhibitor of metalloproteinases 2, KIAA1731NL, anon-WO0134654.19, DmelCG4637, VEGF-A, bar3, DmelCG12249, Mir, MIR, vegf-a, VEGF164, Miranda, MVCD1, Vegf, l(3)neo56, l(3)neo57, MONDOA, Vegf188, DDC8, Timp-2, VPF, Vpf, Mrt, Mira, DANCE, UP50, D11Bwg1104e, ADCL2, Inducible degrader of the LDL-receptor, mir, CSC-21K, Myosin regulatory light chain-interacting protein, VEGF, anon-WO0182946.19, bar-3, MIRA, VEGFA, bHLHe36, vegf, Idol, vegfa, hg</description_synonyms><name_synonyms>Human, Modern., human being, Man (Taxonomy), Homo sapiens, man, Man, human, Modern Man</name_synonyms></additional><is_claimable>false</is_claimable><name>Homo sapiens</name><description>miR-200c is Aberrantly Expressed in Leiomyomas in Ethnic-Dependent Manner and Targets ZEBs, VEGFA, TIMP2 and FBLN5</description><dates><last_updated>2025-09-24</last_updated><first_public>2014-02-11</first_public></dates><accession>PRJNA169086</accession><cross_references><GEO>GSE38817</GEO><taxon>9606</taxon><PubMed>22685266</PubMed></cross_references></HashMap>
