Endothelium comes in close proximity to mast cells and exposed to high concentration of mast cell tryptase that released in substantial quantities at sites of allergic inflammation. Pro-inflammatory actions of tryptase are proposed to be mediated through protease activated receptors 2 (PAR2), but the precise contribution of tryptase to disease processes remains unclear. We have investigated alterations in the whole genome expression profile of endothelial cells in response to tryptase. We used microarrays to detail the changes in the global gene expression profile induced by tryptase or PAR-2 peptide agonist SLIGKV-NH2 in HUVECs and identified distinct classes of up/down-regulated genes and pathways during this process. Overall design: Human umbilical vein endothelial cells (HUVECs) were isolated from umbilical vein tissue and grown to confluence. Following addition of tryptase or the PAR-2 peptide agonist SLIGKV-NH2 or medium alone for 8 hours, total cellular RNA was extracted from HUVEC lysate in three independent experiments. Quantitative polymerase chain reactions (qPCR) was performed and whole genome microarray analysis was performed.</long_description><repository>ENA</repository></additional><is_claimable>false</is_claimable><name>Expression data from HUVECs treated with tryptase or PAR-2 peptide agonist (SLIGKV-NH2)</name><description>Expression data from HUVECs treated with tryptase or PAR-2 peptide agonist (SLIGKV-NH2)</description><dates><last_updated>2021-02-03</last_updated><first_public>2021-02-02</first_public></dates><accession>PRJNA564998</accession><cross_references><GEO>GSE137279</GEO><taxon>9606</taxon></cross_references></HashMap>
