Reprogramming of sorghum proteome in response to sugarcane aphid infestation

Project description:Sugarcane aphids (SCA; Melanaphis sacchari Zehntner) is a key piercing-sucking type pest of sorghum (Sorghum bicolor) which cause significant yield losses. While feeding on host plants, complex signaling networks are invoked from recognition of insect attack to induction of plant defenses. Consequently, these signaling networks lead to the production of insecticidal compounds or limited access to nutrients to insects. Previously, several studies are published on the transcriptomics analysis of sorghum in response to SCA infestation, but no information is available on the physiological changes of sorghum at proteome level. We used SCA resistant sorghum genotype SC265 for the global proteomics analysis after 1 and 7 days of SCA infestation using TMT-plex technique.

Untargeted mutagenesis of brassinosteroid receptor SbBRI1 confers drought tolerance by altering phenylpropanoid metabolism in Sorghum bicolor

Project description:Drought is a critical issue in modern agriculture, therefore there is a need to create crops with drought resilience. The complexity of plant responses to abiotic stresses, particularly in the field of brassinosteroid (BR) signaling, has been the subject of extensive research. In this study, we unveil compelling insights indicating that the BRASSINOSTEROID INSENSITIVE 1 (BRI1) receptor in Arabidopsis and Sorghum plays a critical role as a negative regulator of drought responses. Introducing untargeted mutation in the sorghum BRI1 receptor (SbBRI1) effectively enhances the plant ability to withstand osmotic and drought stress. Through DNA Affinity Purification sequencing (DAP-Seq) we show that the sorghum BRI1-EMS-SUPPRESSOR 1 (SbBES1) transcription factor, a downstream player of the BR signaling, binds to a conserved G-box binding motif, and it is responsible for regulating BR homeostasis, as its Arabidopsis ortholog AtBES1. We further characterized the drought tolerance of sorghum bri1 mutants and decipher SbBES1-mediated regulation of phenylpropanoid pathway. Our findings suggest that SbBRI1 signaling serves as a dual purpose: under normal conditions, it regulates lignin biosynthesis by SbBES1, but during drought conditions, BES1 becomes less active, allowing the activation of the flavonoid pathway. This adaptive shift improves the photosynthetic rate and photoprotection, reinforcing crop adaptation to drought.

Characterization of engineered sorghum with reduced S-adenosyl-L-methionine (AdoMet) levels

Project description:Lignin accumulates progressively in cell walls during plant development, therefore, we are interested in analyzing the transcriptome and metabolome of engineered sorghum lines at three developmental stages. Results from these analyses will help to understand the metabolic changes (if any) besides lignin synthesis in transgenics and anticipate plants' physiological responses during growth under natural environment in future field trials. This project will analyze the transcriptome and metabolome in stems of low-lignin sorghum transgenic lines compared to wild-type (WT) control. Heat stress triggers protein secretion into the extracellular matrix of sorghum cell suspension cultures

Project description:Plants reprogram gene expression as an adaptive response to survive high temperatures. While the identity and functions of intracellular heat stress-responsive proteins have been extensively studied, the heat response of proteins secreted to the extracellular matrix is unknown. Here, we used Sorghum bicolor, a species adapted for growth in hot climates, to investigate the extracellular heat-induced responses. When exposed to 40 C for 72 h, heat-sensitive Arabidopsis cell suspension cultures died, while ICSB338 sorghum cell cultures survived by activation of a transcriptional response characterized by the induction of HSP70 and HSP90 genes. Quantitative proteomic analysis of proteins recovered from cell culture medium revealed specific heat stress-induced protein expression within the sorghum secretome. Of the 265 secreted proteins identified, 100 responded to heat, with the majority (85%) possessing a predicted signal peptide for targeting to the classical secretory pathway. The differentially expressed proteins have putative functions in detoxification (36%), metabolism (32%) and protein modifications (19%), while 13% are unclassified. Extracellular proteins play a vital role in cell-cell communications during stress adaptation. Our study reveals new insights into the adaptive response of sorghum to heat and provides a useful resource of extracellular proteins that could serve as targets for developing thermotolerant crops.
