Project description:Plant hormones and small secretory peptides often function as environmental stress mediators. Some recent reports indicate that small secretory peptides, such as CLAVATA3/EMBRYO SURROUNDING REGION-RELATED (CLE), also function as mediators of environmental stimuli. CLE３ is induced in roots by defense elicitor treatment. Plants without functional CLE3 showed compromized defense gene responses in shoots when plant roots were treated with NaSA. Here, we identified specific genes downstream of CLE3 in roots and shoots with transformed Arabidopsis plants.

Gene expression in response to AGL15 during somatic embryogenesis

Project description:Transcript accumulation was measured using the Affymetrix Arabidopsis ATH1 Genome Array [ATH1-121501] to document changes in response to the MADS-domain transcription factor AGAMOUS-Like 15 during somatic embryogenesis. A somatic embryo system was used where mature seed is allowed to complete germination in liquid MS media containing 2,4-D and seedlings produce somatic embryos from the shoot apical meristem (SAM) region. The frequency with which these embryos are produced directly correlates with AGL15 accumulation. Experiment Overall Design: Wild type Arabidopsis (Columbia ecotype) with normal amounts of AGL15 were compared to (1) 35S:AGL15 that constitutively expresses AGL15 and produces more somatic embryos from the shoot apical region than wild type and (2) agl15 agl18 double loss-of-function mutants that produce less somatic embryos from the shoot apical region than wild type.

Different gene expressions in root tips between CLE10 and CLE25 treatment

Project description:Cell–cell communication is critical for tissue and organ development. In plants, secretory CLAVATA3/EMBRYO SURROUNDING REGION-related (CLE) peptides function as intercellular signaling molecules in various aspects of tissue development. However, little is known about intracellular signaling pathways functioning in vascular development downstream of the CLE ligands. To elucidate CLE signaling pathway, we performed GeneChip analysis. Arabidopsis roots were treated with chemically synthesized CLE10, or CLE25 at 1000 nM for 12 hours. Comparisons between CLE10 and CLE25 treatment will highlight the genes involved in the cause of different effects.

Expression data from Arabidopsis Col, C24 and their F1 hybrid at 15 DAS

Project description:To obtain new insights for heterosis mechanisms in Arabidopsis, transcript profiling of arabidopsis F1 hybrid and their parentral lines was obtained from Affymetrix GeneChips ATH array. The microarray analysis exhibited that there were 44 up-regulated and 12 down-regulated genes with more than a 1.5-fold changes in the profiles of F1 hybrid against those of each parent. Gene ontology analyses highlighted up-regulated genes as organic nitrogen-related in the F1 hybrid.
