Project description:Clinical Significance: Understanding the differences in colorectal cancer (CRC) aggressiveness and clinical outcomes in relation to tumor stage and different molecular subsets is at most important for designing treatment regimens. However, molecular signatures for specific phenotypic subsets that predict the aggressiveness and clinical outcomes of CRC, specifically in advanced disease stage are lacking. Therefore, for the first time, the current study has identified a set of molecular markers that are associated with aggressive Stage III CRCs that exhibited microsatellite stable and mutant p53 phenotypic features. These findings might aid in designing aggressive treatment regimens and help to provide insights into the development of novel therapeutic targets. Results: Increased incidence of p53 mutations in MSS CRCs (58%) was associated with higher CRC-specific mortality than MSS-p53 wild-type phenotypes (log-rank, P=0.025; and hazard ratio, 2.52; 95% confidence interval, 1.25-5.08). Of 49 down-regulated genes, LPAR6, PDLIM3 and PLAT and of 35 up-regulated genes, TRIM29, FUT3, IQGAP3, and SLC6A8, were confirmed by qNPA, qRT-PCR, and IHC platforms. Project description:We evaluated the profile of miRNA expression in 6 colorectal adenoma (CRA), 6 colorectal adenocarcinoma (CRC) and 6 matched normal mucosa (NOR) using the Exiqon miRCURY LNA microRNA array,7th generation. We found that global dysregulated miRNAs between colorectal lesions and normal mucosa. Our findings implicates that dysregulation of miRNAs may play important role in the carcinogenesis and present therapeutic targets for CRC.

Project description:Abstract: Altered expression of microRNAs (miRNAs) is associated with development and progression of various human cancers by regulating the translation of oncogenes and tumor suppressor genes. In colorectal cancer (CRC), these regulators complement the Vogelstein multi-step model of pathogenesis and have the potential of becoming a novel class of tumor biomarkers and therapeutic targets. Using QRT-PCR, we measured the expression of 621 mature miRNAs in 40 CRCs and their paired normal tissues and identified 23 significantly deregulated miRNAs. We subsequently evaluated their association with clinical characteristics of the samples and presence of alterations in the molecular markers of CRC progression. Expression levels of miR-31 were correlated with CA19-9 and miR-18a, miR-21 and miR-31 were associated with mutations in APC gene. To investigate the downstream regulation of the differentially expressed miRNAs identified we integrated putative mRNA target predictions with the results of a meta-analysis of seven public gene expression datasets of normal and tumor samples of CRC patients. Many of the CRC deregulated miRNAs computationally map to targets involved in pathways related to progression. Here one promising candidate pair (miR-1 and MET) was studied and functionally validated. We show that miR-1 can have a tumor suppressor function in CRC by directly down-regulating MET oncogene both at RNA and protein level and that re-expression of miR-1 leads to MET driven reduction of cell proliferation and motility, identifying the miR-1 down-modulation as one of the events that could enhance CRC progression. Project description:Colorectal cancer (CRC) poses one of the most serious threat against human health worldwide, and abnormally expressed c-Myc and p53 are deemed the pivotal driving forces of CRC progression. Here we discovered a lncRNA FIT, which was down-regulated in CRC clinical samples, was transcriptionally suppressed by c-Myc and promoted CRC cells apoptosis by inducing FAS expression. FAS is a p53 target gene, and we found that FIT formed a trimer with RBBP7 and p53 which facilitated p53 acetylation and transcription. Moreover, FIT was capable of retarding CRC growth in mice xenograft model and positively relevant to FAS expression clinically. Thereby our study elucidated the role of lncRNA FIT in human colorectal cancer growth and provides a potential target for anti-CRC drugs.

Project description:Defining molecular features that can predict the response to chemotherapy for stage II-III colorectal cancer (CRC) patients remains challenging in cancer research. Most available clinical samples are Formalin-Fixed and Paraffin-Embedded (FFPE). Affymetrix GeneChip® Human Transcriptome Array 2.0 (HTA) is one platform marketed for high-throughput gene expression profiling for FFPE tissue samples. In this study, we analyzed the whole transcriptom gene expression of 156 CRC patient samples measured by this platform to identify biomarkers predicting the response to chemotherapy for stage II-III CRC patients.

Project description:In many tumors, the tumor suppressor TP53 (p53) is not mutated, but experiences functional inactivation. Notwithstanding, restoring p53 function emerges as an appealing therapeutic approach for tumors exhibiting wild-type p53 (p53WT). However, the mechanisms underlying the functional inactivation of p53 in these tumors remain poorly understood. Previously, we identified SETD8 as a critical suppressor of p53 activity in neuroblastomas. SETD8 is the sole known enzyme that mono-methylates p53 on lysine 382 (p53K382me1), resulting in the inhibition of its pro-apoptotic and growth-arresting functions. In this study, we observed high expression levels of SETD8 and p53K382me1 in 185 colorectal cancer (CRC) tissue samples, being negatively associated with patient survival probability. Significantly, the expression of p53K382me1 resulted confined to colorectal cancer stem cells and tumor-associated macrophages (TAM) in tissues obtained from CRC patients. Within CRC patient cohorts, high expression of p53K382me1 levels in both stem and immune cells predict decreased survival probability. The SETD8-mediated inactivation of p53 through p53K382me1 may serve as an early requirement for tumor initiation, particularly in inflammation-induced cancers, and as a functional biomarker and therapeutic target in advanced CRCs

Project description:Colorectal cancer (CRC) progresses through distinct molecular mechanisms, tumor stage playing a pivotal role in prognosis and treatment outcomes. Despite advances in CRC treatment, the lack of stage-specific biomarkers limits the potential for reliable diagnostic and treatment strategies. This study aims to identify stage-specific gene expression profiles and the molecular mechanisms through comparative transcriptomic analysis of adenoma (n = 10), carcinoma in situ (n = 8) and adenocarcinoma (n = 11) samples. Utilizing RNA-seq from FFPE samples processed with absGSEA, we identified significant cellular pathways influenced from each CRC stage. In adenocarcinoma, pathways related to transcriptional co-regulatory mechanisms and protein kinase functional activity, critical for tumor growth and metastasis, were significantly enriched compared to adenoma. While apoptotic processes and Wnt signaling pathways were prominent in carcinoma in situ compared to adenoma samples. Extracellular matrix organization pathway was significantly enriched between adenocarcinoma and carcinoma in situ. Transcriptomics data and bioinformatics analysis revealed that in adenocarcinoma cohorts, genes such as COL1A2, CEBPZ, MED10, and PAWR were overexpressed, highlighting their potential roles in advanced disease progression. Conversely, in adenoma samples, the overexpression of genes such as ARRB1, CTBP1, and CTBP2 underscores their involvement in early tumorigenesis. COL1A2 and CEBPZ were implicated in extracellular matrix remodeling and transcriptional regulation respectively, while and ARRB1 were associated with early tumorigenesis. These findings highlight the complexity of gene regulation in CRC and provide a foundation for developing stage-specific diagnostic and prognostic biomarkers, and to ultimately improve therapeutic strategies.
