Project description:The Hypoxia-Inducible Factors induce the expression of the histone demethylases JMJD1A (KDM3A) and JMJD2B (KDM4B), linking the hypoxic tumor microenvironment to epigenetic mechanisms that may foster tumor progression. Using transcript profiling, we have identified genes that are regulated by JMJD1A and JMJD2B in both normoxic and hypoxic conditions in SKOV3ip.1 ovarian cancer cells. This dataset includes expression data obtained from exposing ovarian cancer cells to hypoxia in combination with siRNA-mediated knockdown of the hypoxia-inducible histone demethylases JMJD1A and JMJD2B. These data were used to both identify functional overlap between each histone demethylase, as well as identify effectors of tumor growth mediated by JMJD2B (KDM4B) in normoxia and hypoxia.
Project description:Profiling of changes in steady state RNA levels upon RNAi-mediated knockdown of the nucleosome remodeling ATPase ISWI in Drosophila SL2 cells. Quantitation of the corresponding proteomes has been performed to describe transcriptome-proteome relations. The analysis indicates a limited correlation of the two expression steps. Keywords: RNAi-mediated knockdown

Project description:MM1S cells have been cultured under normoxic and hypoxic conditions, and gene expression profiling has been performed using the Affymetrix Human Genome U133 Plus 2.0 array.
Project description:RNAi mediated depletion of TCF7L1 increases activity of a Wnt-based reporter and imparts more aggressive tumor phenotypes in vitro in pancreatic cancer cell lines that express TCF7L1. We sought to determine what changes in transcription ocurred after RNAi mediated knockdown of TCF7L1 in these cells by RNA-sequencing
Project description:HRE1 and HRE2 are two ERF transcription factors induced by low oxygen. In this work we analyzed the effect of ectopic expression of HRE1 and HRE2 on the arabidopsis transcriptome in aerobic and hypoxic (1% O2) conditions. While HRE1 has a moderate effect on the expression of anaerobic genes under hypoxia, HRE2 does not affect them either under aerobic or hypoxic conditions.
Project description:Arbuscular mycorrhiza (AM) interactions between plants and Glomeromycota fungi primarily support phosphate aquisition by the host. To unravel the role of the AM-specific GRAS transcription factors MtGras1 and MtRam1, we performed genome-wide transcriptome profiling in RNAi-mediated MtGras1 knockdown and MtRam1 (ram1-1) mutant roots. Specifically, we used GeneChip Medicago Transcriptome Assays to identify genes differentially regulated in mycorrhizal MtGras1 knockdown or MtRam1 (ram1-1) mutant roots in comparison to control roots.
Project description:Aspergillus fumigatus strains CEA10 and srbA mutant expression patterns were compared after exposure to 24h of hypoxic conditions Goal of this study was to find genes that are directly and/or indirectly regulated by the transcription factor srbA, that is know to be crucial for growth under hypoxic conditions Keywords: hypoxic stress response two strains, 2 biological replicates with dye-swaps
