ABNORMAL POLLEN 1 (AP1) encodes a legume-type lectin receptor-like kinase and is involved in starch accumulation during rice pollen maturation

Project description:Starch accumulation is the key progress for the maturity of rice pollen grains. However, the regulatory mechanism underlying which remains less understood. Here, we isolated a rice male-sterile mutant ap1, which produces non-viable pollen grains with defective starch accumulation. Functional analysis revealed that AP1 encodes an active L-type lectin receptor-like kinase (L-LecRLK). AP1 is located on plasma membrane and its transcript is highly accumulated in pollen during the starch synthesis phase. RNA-seq analysis suggests that the mutation of AP1 significantly altered the expression of numerous genes involved in starch and sucrose metabolic pathway. Phosphoproteomic profiling revealed that the phosphorylation levels of several proteins within this pathway were significantly downregulated in the mutant. Among which, a rice UDP-glucose pyrophosphorylase (OsUGP2), which roles in pollen starch accumulation, was identified. Further protein–protein interaction assays suggest that OsUGP2 is a feasible target of AP1 to regulate pollen starch accumulation. Our findings thus revealed a novel role of L-LecRLK in controlling pollen maturity via modulating starch metabolism.

Project description:In this study, integrated transcriptomics, proteomics and metabolomics approaches were applied to investigate the molecular responses of O3 in the leaves of two-weeks old rice (cv. Nipponbare) seedlings exposed to 0.2 ppm O3 for a period of 24 h. Based on the morphological alteration of O3-exposed rice leaves, transcript profiling of rice genes was performed in leaves exposed for 1, 12 and 24 h using rice DNA microarray chip, proteomics and metabolomics. This systematic survey showed that O3 triggers a chain reaction of altered gene, protein and metabolite expressions involved in multiple cellular processes in rice. Also investigated were the molecular responses in the leaves of two-weeks old rice (cv. Nipponbare) seedlings under continuous light and pure air (as a positive control for ozone exposure experiments) for a period of 24 h. Transcript profiling of rice genes was performed in leaves exposed for 1, 12 and 24 h using rice DNA microarray chip, proteomics and metabolomics. This systematic survey showed that continuous light and growth for 24 h also triggers a chain reaction of altered gene expressions involved in multiple cellular processes in rice, but different from those against ozone, in general. Global expression analysis of differential expression of genes working under Soil Water Stress condition in different genotypes of rice in Vegetative and Grain-filling stage [set 1]

Project description:One of the serious constraints to realize high level of rice crop productivity in Indian agriculture has been due to soil moisture stress (SMS) situation that growing plants often face. In order to increase or maintain the crop productivity in SMS situation our initial aim is to understand the drought response mechanism in different genotypes of rice. For thorough analysis of SMS situation in rice we have taken here two wild genotypes of rice namely Oryza nivara, Oryza rufipogan and three Indian cultivar namely Oryza Nagina-22, Oryza IR20 and Oryza Vandana, where IR20 is known to be susceptible and Vandana is known to be tolerant under SMS condition. Global analysis of transcript profiling under SMS condition reveal the actual picture of genes responsive to stress situation in different genetic background of rice. Furthermore it would help us in the selection of most desirable resource for crop breeding without compromising the yield of crop. We used the 44k rice Oligoarray from Agilent technologies to study the transcript profiling from five genotypes of rice under control, soil moisture stress and after recovery conditions during vegetative and grainfilling phase.Here in case of Nagina-22 we have taken grainfilling stage.
