Project description:Systemic acquired resistance (SAR) is a plant defense response that provides long-lasting, broad-spectrum pathogen resistance to uninfected distal leaves following an initial localized infection. However, little information is available on the molecular biological basis of SAR especially in uninfected distal leaves. Here, we used two SAR-induced bacteria to induce SAR in Arabidopsis, a metabolomics approach based on ultra-high-performance liquid chromatography (UPLC) and mass spectrometry (MS) technique was used to identify SAR-related metabolites in SAR-induced bacteria infected local leaves and distal leaves of locally SAR-induced bacteria infected plants. Multiple statistical analyses were used to identify differentially expressed metabolites (DEMs). The result showed primary metabolism and secondary metabolism were significantly altered in local leaves and distal leaves, including phenolic compounds, amino acids, nucleotides, organic acids and many other metabolites. Furthermore, the content of amino acids, phenolic compounds, coenzymes and its derivatives increased in distal leaves, suggesting these metabolites contributed to the establishment of SAR. In addition, 2’-hydroxy-4,4’,6’-trimethoxychalcone, phenylalanine and ethyl docosahexaenoate were identified as potential components which may play important role both in basic resistance and SAR. This study provided a reference for understanding metabolic mechanism associated with SAR in Arabidopsis, which will be useful for further investigation of the molecular basis of SAR.

Project description:The epidermis of plants forms a protective barrier against biotic and abiotic stress. Little is known about how breaches in the epidermis are repaired, especially those of mature leaves. Here, we investigated wound healing in the mature leaves of Arabidopsis. We discover a novel wound protection mechanism comprising a multi-layered ligno-suberized barrier covered with cuticular wax. This barrier is formed by mesophyll cells that adopt an epidermal fate. This cell fate transition is regulated in two steps by ATML1, a key transcription factor in epidermal specification. First, mesophyll cells of protective layer 1 (P1), just beneath the wound, transition into epidermal cells, sealing the wound by depositing cuticle, a mechanism that involves signaling through ethylene and reactive oxygen species produced by RBOHE. This signaling also promotes P1 cell death, ensuring wax accumulation on the surface. Second, the underlying protective layer 2 (P2) undergoes ligno-suberization, driven by jasmonic acid and RBOHD, forming a cork-like layer on the leaf surface. ATML1 regulates this process in P2 as well. Thus, wound healing in mature leaves involves integration of ethylene and jasmonic acid signaling with ATML1-mediated epidermal cell specification to coordinate cell layer-specific functions, including cuticular wax formation and ligno-suberization. This novel protective mechanism also occurs in the leaves of tobacco and Capsella, suggesting it is widespread.

Project description:Wounding due to mechanical injury or insect feeding causes a wide array of damage to the cell including cell disruption, desiccation, metabolite oxidation, and disruption of primary metabolism. In response, plants regulate a variety of genes and metabolic pathways to cope with injury. Tomato (Solanum lycopersicum) is a model for wound signaling but few studies have examined the comprehensive gene expression profiles in response to injury. A cross-species microarray approach using the TIGR potato 10-K cDNA array was analyzed for large-scale temporal (early and late) and spatial (locally and systemically) responses to mechanical wounding in tomato leaves. These analyses demonstrated that tomato regulates many primary and secondary metabolic pathways and this regulation is dependent on both timing and location. To determine if LAP-A, a known modulator of wound signaling, influences gene expression beyond the core of late wound-response genes, changes in RNAs from healthy and wounded Leucine aminopeptidase A-silenced (LapA-SI) and wild-type (WT) leaves were examined. While most of the changes in gene expression after wounding in LapA-SI leaves were similar to WT, overall responses were delayed in the LapA-SI leaves. Moreover, two pathogenesis-related 1 (PR-1c and PR-1a2) and two dehydrin (TAS14 and Dhn3) genes were negatively regulated by LAP-A. Collectively, this study has shown that tomato wound responses are complex and that LAP-A's role in modulation of wound responses extends beyond the well-described late-wound gene core.
