Transcriptome profiling of Hu sheep testicular development associated with dietary energy restriction and during compensatory compensation

Project description:To investigate the effect of energy levels on sheep testicular development, we systematically investigated mRNAs using high-throughput sequencing at two dietary energy levels of Hu sheep (maintenance energy and 55% energy levels) chrysophanol alone treatment and combination treatment as well as revealed that the combination treatment the most defence-related genes induced expression changes. We identified numerous differentially expressed genes that exhibited distinct expression patterns, and defence-related genes that were highly differentiated between the physcion and chrysophanol alone treatment and combination treatment. These genes have known or potential roles in defense response and were enriched in functional gene categories potentially responsible for cucumber resistance. Therefore, they are appealing candidates for further investigation of the gene expression and associated regulatory mechanisms related to defense response.

Next Generation Sequencing Facilitates Quantitative Analysis of Physcion and Chrysophanol on cucumber powdery mildew Transcriptomes

Project description:We generated 17.6 Gb of high-quality sequencing data (~2 Gb per sample) and catalogued the expression profiles of 12,293 annotated cucumber genes in each sample. The analysis showed differences of transcriptomes between physcion and chrysophanol alone treatment and combination treatment as well as revealed that the combination treatment the most defence-related genes induced expression changes. We identified numerous differentially expressed genes that exhibited distinct expression patterns, and defence-related genes that were highly differentiated between the physcion and chrysophanol alone treatment and combination treatment. Dual inhibition of SHP2 and autophagy suppresses NF1-associated malignant peripheral nerve sheath tumors

Project description:Malignant peripheral nerve sheath tumors (MPNSTs) are the leading cause of premature death for patients with Neurofibromatosis type 1 and no approved targeted therapies are available. Benign plexiform neurofibromas have been successfully treated with selumetinib, a MEK inhibitor, but after progression to MPNST, MEK inhibition alone is not effective. Frequently, adaptive responses to single agent targeted inhibitors invokes alterations in receptor tyrosine kinase expression and feedback regulation that leads to inhibitor bypass. Here, the effects of SHP099, an inhibitor of the protein-tyrosine phosphatase SHP2 (encoded by the PTPN11 gene) was tested alone and in combination with trametinib (MEKi) in an NF1 MPNST patient derived xenograft model (PDX4) treated with SHP099, hydroxychloroquine (HCQ) or the combination. Kinase enrichment proteomic analysis was performed using tumor tissue treated with SHP099, hydroxychloroquine (HCQ) or the combination to evaluate the effects on the functional kinome.

Effect of bromodomain inhibitors on dopamine D1R induced gene expression in striatal neurons

Project description:The dopamine D1R is Gs coupled GPCR which is expressed in striatal neurons and stimulates gene expression upon activation by dopamine or other agonists. In this work we investigated the role of the BET protein bromodomain containing protein 4 (Brd4) in mediating D1R-dependent gene expression in rat striatal neurons. Here we report the results of 3 RNA sequencing experiments. In Experiment 1 we treated primary striatal neurons with the D1R-selective agonist SKF-81297 alone or in combination with the non-selective BET inhibitor JQ1 for 60 minutes to assess the effect of BET inhibition on acute D1R-dependent gene expression. In Experiment 2, we treated primary striatal neurons with the D1R-selective agonist SKF-81297 alone or in combination with the BET bromodomain 2 selective inhibitor iBD2 for 60 minutes to determine the contribution of BD2 specifically to D1R-dependent gene expression. In Experiment 3 we treated primary striatal neurons with the D1R-selective agonist SKF-81297 alone or in combination with the non-selective BET inhibitor JQ1 for 24 hours to assess the effect of prolonged BET inhibition on basal gene expression and D1R-dependent gene expression.
