Characterization of CpeNeo mice, a conditional null mutation of carboxypeptidase E, and restoration of Cpe expression with Flp recombinase

Project description:Carboxypeptidase E (CPE) is an essential enzyme that contributes to the biosynthesis of the vast majority of neuropeptides and peptide hormones. Mice lacking CPE activity due either to a natural mutation or targeted disruption of the Cpe gene have greatly decreased levels of most neuropeptides and show a large number of defects including severely impaired fertility, adult-onset obesity, anxiety- and depressive-like behavior, and abnormal hippocampal development. In the present study, we generated a mouse line named CpeNeo due to a neomycin cassette within the Cpe gene that blocks normal splicing of the gene, leading to the loss of enzyme expression. CpeNeo/Neo mice show all of the defects previously found with Cpefat/fat and/or KO mice. In addition, young adult CpeNeo/Neo mice are more sensitive to morphine-induced hyperlocomotion than wild-type mice, while older CpeNeo/Neo mice are less sensitive than wild-type mice. Removal of the neomycin cassette with Flp recombinase under a germline promoter restored the expression of CPE activity, leading to normal levels of neuropeptides in brain. CpeFlp/Flp mice are identical to wild-type mice in terms of behavior and physiology. Mice heterozygous for the CpeNeo allele have Cpe mRNA levels ~50% of wild-type mice, but are identical to wild-type mice in all behaviors and physiological parameters examined. These results confirm that CPE is not a rate-limiting enzyme in the production of most neuropeptides, despite claims that small decreases in CPE activity contribute to obesity or other physiological effects

Transcription profiling of mouse spontaneous liver tumors of Trim24/TIF1a-null animals

Project description:The transcriptional coregulator Trim24 (formerly known as TIF1a) functions in mice as a liver-specific tumor suppressor. Mice carrying a null mutation in the Trim24 gene all develop hepatocellular carcinoma (HCC). We used microarrays to identify the alterations in gene expression patterns associated with loss of Trim24 and consequent HCC development. Experiment Overall Design: Five independent liver tumor (HCC) samples taken from five Trim24 knockout (KO) mice and five normal liver tissue samples taken from wild-type (WT) littermate controls were used for RNA extraction and hybridization on Affymetrix microarrays.

LKB1 and Notch deletion in mouse liver

Project description:We characterize the phenotype of mice in which the deletion of Lkb1 has been targeted in the liver. Lack of Lkb1 in the liver results in bile duct paucity leading to cholestasis. This phenotype is similar to that obtained upon inactivation of Notch signaling in the liver. We test the hypothesis of a functional overlap between the Lkb1 and Notch pathways by gene expression profiling of livers deficent in Lkb1 or in the Notch mediator RbpJÎº. We used AlfpCre mice for liver-specific deletion of LKB1 that were crossed with a conditional knockout mouse model (LKB1 floxed mice). We used also AlfpCre mice for liver-specific inactivation of the Notch pathway. AlfpCre mice were crossed with the RbpJÎº floxed mice. RNA was extracted from the liver of LKB1 KO mice (Lkb1Floxed, AlfpCre positive mice) and their wild-type counterpart (Lkb1Floxed, AlfpCre negative mice). RNA was also extracted from liver of RbpJK KO mice (RbpJK floxed, AlfpCre positive) and their wild type mice (RbpJK floxed, AlfpCe negative). 6 samples for the liver-specific deletion of LKB1 corresponding to 3 KO LKB1 (Cre positive) and 3 normal liver (Cre negative). 6 samples for the liver-specific inactivation of the Notch pathway corresponding to 3 KO RbpJk (Cre positive) and 3 normal liver (Cre negative).

RNA-Seq Reveals Enhanced Inflammation and Fibrosis in Alcohol-Induced Liver Injury of Apoe Knockout Mice

Project description:Alcohol-related liver disease (ALD) involves inflammation, hepatocellular damage, and fibrosis. Apolipoprotein E (Apoe) plays a role in immune regulation and tissue repair, but its function in ALD remains unclear. To explore this, we performed RNA sequencing on liver tissues from wild-type (WT) and Apoe knockout (Apoe KO) mice subjected to a chronic-plus-binge ethanol model. Gene expression analysis revealed distinct clustering between groups and significant alterations in pathways related to neutrophil degranulation, extracellular matrix organization, and collagen formation, indicating heightened inflammation and fibrosis in Apoe KO mice. These findings offer insights into Apoe’s role in ALD and liver repair.

Proteomic and phosphoproteomic analysis of PKCe KO

Project description:Activation of protein kinase C epsilon (PKCε) in the liver has been widely associated with hepatic insulin resistance. PKCε is proposed to inhibit insulin signalling through phosphorylation of the insulin receptor. We have tested this directly by breeding PKCε floxed mice with mice expressing Cre recombinase under the control of the cytomegalovirus, albumin or adiponectin promoters to generate global, liver- and adipose tissue-specific PKCε knockout (KO) mice. Global deletion of PKCε recapitulated the benefits for diet-induced glucose intolerance that we previously described using conventional PKCε KO mice. However, we did not detect PKCε-dependent alterations in hepatic insulin receptor phosphorylation. Furthermore, liver-specific KO mice were not protected against diet-induced glucose intolerance or insulin resistance determined by euglycemic clamp. In contrast, adipose tissue-specific KO mice exhibited improved glucose tolerance and mildly increased hepatic triglyceride storage, but no change in liver insulin sensitivity. Phosphoproteomic analysis of insulin signalling in PKCε KO adipocytes revealed no defect in the canonical INSR/AKT/mTOR pathways. However, PKCε KO resulted in changes in phosphorylation of several proteins associated with the endosome and cell junctions suggesting regulation in secretory pathways and a potential role of PKCε in endocrine function. Indeed, RNA-seq analysis revealed adipose-tissue PKCε-dependent changes in the hepatic expression of several genes linked to glucose homeostasis and hepatic lipid metabolism. The primary effect of PKCε on glucose homeostasis is therefore not exerted directly in the liver as currently assumed. However, PKCε in adipose tissue modulates glucose tolerance and is involved in crosstalk with the liver that affects gene expression and lipid accumulation.
