Parasites expressing GFP-fused PbSIP2 (PbSIP2::GFP) in cultures were harvested at 24 h post infection and subjected to ChIP-seq experiments using anti-GFP antibody. Binding sites of PbSIP2 were determined from the sequence data.

RNA-Seq of Plasmodium chabaudi AS erythrocytic stages

Project description:Recent advances in high throughput sequencing methodologies allow the opportunity to probe in depth the transcriptomes of organisms including important human pathogens. In this project, we are using Illumina sequencing technology to analyze the transcriptome (RNA-Seq) of experimentally accessible stages of the mouse malaria parasite, P. chabaudi AS. The aim is to analyse cir gene expression during Plasmodium chabaudi infection and determine whether host genetic background can influence cir expression. This data is part of a pre-publication release. m5C methyltransferase NSUN3 regulates variant gene expression and development in blood-stage Plasmodium falciparum [RNA-seq]

Project description:C5 methylation of cytosine is a relatively abundant post-transcriptional modification in eukaryotes, and the NSUN family has been identified as an important executor of m5C modification. Currently, little is known about this family in Plasmodium spp. Therefore, we constructed a pfnsun3 gene knockdown strain and the knockdown efficiency was confirmed through growth curves and western blot experiments. The target genes of PfNSUN3 was obtained by RNA immunoprecipitation and high-throughput transcriptome sequencing experiments. Our data reveal that pfnsun3 is an indispensable post-transcriptional RNA modification in regulating variant gene expression in Plasmodium falciparum.
