Gene Expression in wild type and CCR5 knockout mice with lung metastasis

Project description:We have shown that C57BL/6J CCR5 knockout mice develop 30.4% ± 8.6% fewer B16 F10 lung nodules compared to wild type mice after the intravenous injection of 100,000 B16 F10 cells. We sought to understand this phenomenon by comparing gene expression in the lungs of these mice at 6, 24, and 48 hours after tumor injection.

RNA sequencing analysis of busulfan-treated initial segment-caput epididymis.

Project description:Purpose: The purpose of this study is to characterize genes whose expressions in the initial segment (IS)-caput epididymis were affected by busulfan intraperitoneal injection. Methods: 8-week-old wild-type mice were treated with single intraperitoneal injection of dimethyl sulfoxide (DMSO) or busulfan (40mg/kg body weight) disolved in DMSO and mice were left for 4 weeks until tissue sampling at 12 weeks old. IS-caput epididymal mRNA profiles were generated by deep sequencing, in triplicate, using Illumina NovaSeq6000. Results: RNA-seq data identified differentially expressed transcripts. Conclusions: Our results show that the expression of many genes was affected by busulfan injection.

Acute lung injury models identify putative growth factors that promote tissue repair and biomarkers for acute lung injury

Project description:We tried to identify growth factor(s) to promote regeneration of type 2 alveolar epitherial cells. To identify such candidates, we compared gene expression profiles of RNAs from lungs of mice of the following four different groups of bone marrow (BM) chimeric mice after diphteria toxin (DT) injection. LL, BM cells of LysM-DTR mice into LysM-DTR mice; WL, BM cells of wild-type mice into LysM-DTR mice; LW, BM cells of LysM-DTR mice into wild-type mice; WW, BM cells of wild-type mice into wild-type mice. Transcriptome analysis revealed that a set of growth factor-related genes was upregulated in the lungs of WL mice compared to LL mice.
